The Mechanism Study Of Xuanbi Decoction On Anti-inflammatory And Analgesic Effects Of Rheumatoid Arthritis Model In Mouse

Purpose: Observe the mechanism study of Xuanbi Decoction on anti-inflammatory and analgesic effects through establish the mice model of Rheumatoid Arthritis,and record the change about the volume of the toes,the WBC in peripheral blood leucocyte,and the cytokines of IL-1??IL-4?IL-6?TNF-??INF-?,all these can provide the experimental basis to prove that Xuanbi Decoction has a definite therapeutical effect on creat RA.Methods: 100 mice were randomly divided into five groups,model group,western medicine control group,Xuanbi Decoction high,medium and low dose group,20 rats in each group.The analgesic effect of Xuanbi Decoction was observed by mice acetic acid twisting test,and the anti-inflammatory mechanism of Xuanbi Decoctionwas observed by mice ear edema which was caused by dimethylbenzene.60 mice were randomly divided into six groups,blank control group,model group,western medicine control group,Xuanbi Decoction high,medium and low dose group,10 rats in each group.Through the establishment of collagen induced arthritis model,to observe the effect of Xuanbi Decoction of arthritis model mouse foot swelling effect and the change of the cytoline on IL-1??IL-6?TNF-??IL-4?IFN-?.Results: Xuanbi Decoction high,medium and low dose group can reduce the frequency of acetic acid twisting,compared with the control group,the difference is significant(P<0.05).Xuanbi Decoction can ease the extent in mice ear edema ? Xuanbi Decoction high,medium and low dose group can ease the collagen of joint inflammation in mice hind paw edema,compared with the modell group,the difference is significant(P<0.05).Xuanbi Decoction high dose group can ease the content of peripheral blood leucocyte,compared with the modell group,the difference is significant(P<0.05).Xuanbi Decoction high,medium and low dose group have an obvious inhibiting effect on IL-1??TNF;compared with the modell group,the difference is significant.Xuanbi Decoction high dose group can ease the content of IL-6;THE experimental groups ' cytokines of IFN-? in peripheral blood have a lower expression,compared with the modell group,the difference is significant(P<0.01),compared with the blank control group,the difference is inapparent(P>0.05);THE cytokines of IL-4 in peripheral blood of experimental groups have a higer expression,compared with the modellgroup,the difference is significant(P<0.01),compared with the blank control group,the difference is inapparent(P>0.05);Compared with the modell group,the spleen weight index of mices on Xuanbi Decoction high,medium and low dose group decline significantly(P<0.05),compared in the drug group,the difference is inapparent.Conclusion: Xuanbi Decoction has a better therapeutic effect on anti-inflammatory and analgesic effects.
