The Study Of Jia-wei-qing-hao-bie-jia Decoction Intervening In The Process Of Apoptosis On Lewis Lung Carcinoma Mouse Models

PurPose : Using Jia-Wei-Qing-Hao-Bie-Jia Decoction intervened in lewis lung carcinoma mouse models to research on the mouse models being influenced by the decoction,through observing the common state and tumour's volume and weight and tumour pathological change of the mouse models,and then to research on the effect of the decoction in the process of apoptosis of lewis lung carcinoma.Method :(1)Lewis lung carcinoma mouse models were established by injecting C57BL/6 mice with LLC cells at subcutaneous tissue of the right posterior axillary line.Depending on the size of the tumour,the mice were randomLy divided into the model group,the high dosage group of Jia-Wei-QingHao-Bie-Jia Decoction,the middle dosage group of Jia-Wei-Qing-Hao-Bie-Jia Decoction,the low dosage group of Jia-Wei-Qing-Hao-Bie-Jia Decoction and the cisplatin group,when the Lewis lung carcinoma mouse models were successfully established.(2)The tumour size and weight of the mice were measured and recorded.At the same time,we needed to observe general living conditions such as the colour and gloss of mouse hair,the state of behavior and spirit,etc.(3)The changes of pathological morphology of tumour are observed,through some method of pathological examination such as H-E staining.(4)The expression of Bax protein and gene is detected,using the means of immunohistochemistry and reverse transcription PCR.Results:(1)From the 12 th day after the day that the mold was made,some states emerged into each group such as dull-looking,the hair that becames sparse,sere and lusterless,less activity,like crouch,inappetence,marasmus and becoming insensitivity etc.Especially in the cisplatin group,these states were more palpable and in Chinese medicine group,those symptoms were decreased.(2)The tumour size and weight of the mice in each group were measured,and the results shows that the drug group can significantly inhibit tumorous growth.Comparing with the model group,the difference is significant.(3)Under optical microscope,the observation results of H-E staining of the tumour tissues shows.that apoptosis significantly emerge out of the tumor cells.Comparing with the model group,the difference is significant.(4)The observation results of the expression level of Bax protein and gene shows that the drug group can facilitate the expression of Bax protein and gene.Comparing with the model group,the difference is significant.Conclusion : Jia-Wei-Qing-Hao-Bie-Jia Decoction can facilitate apoptosis of tumour cells to inhibit tumorous growth,by boosting the expression of Bax protein and gene,and can improve the organism's living state.
