The Effects Of Qing Re Hua Tan Tong Fu Decoction To IL-6,VIP And SP Of Brain,Lung,Large Intestine Tissues In Rats With Cerebral Infarction Complicated With Bacterial Pneumonia

Objectives:To establish a rat model of cerebral infarction complicated with bacterial pneumonia,and to observe the effect of Qing Re Hua Tan Tong Fu Decoction on the expression of IL-6,VIP and SP in the brain,lung and large intestine of model rats.Methods:The SPF level of 30 male SD rats were randomly divided into normal group(A group),cerebral infarction complicated with pneumonia model group(B group),low dose drug intervention group(C group),middle dose drug intervention group(D group),high dose drug intervention group(E group).5groups in all,6 rats in each group.The rats in group A without any treatment;Rats in B group,C group,D group,E group were established the model of cerebral infarction with bacterial pneumonia by using modified thread embolism method with trachea injection method.After the establishment of the model,B group were given saline enema,C group,D group,E group were treated with low dose,middle dose,high dose of liquid medicine for rectal administration,1times per day,continuous administration of 7 days.After the establishment of the model,measured the body temperature and weight everyday;In 7~thh day,rats were observed by gross,active,nasal secretions,defecation and neurological deficit score.Also observe pathological condition of brain,lung and large intestine tissues by HE staining,and measure IL-6 expression in brain,lung and large intestine tissues by immunohistochemistry,test the content of VIP and SP of brain,lung and large intestine tissues and serum by ELISA.Results:1.General state:After modeling,rats in B group were generally poor state,slow action,eating and drinking reduction,weight loss,reduce the amount of defecation,also appeared sneezing and respiratory dyspnea aggravated,nasal secretions increased,temperature rise higher.Rats in C group,D group,E group were better than the B group in the general state,body temperature,and body weight(P<0.05).2.The neurological impairment score:The neurological impairment score of rats in B group was significantly lower than those in A group(P<0.01).The neurological impairment score of rats in E group was significantly higher than those in B group(P<0.01).3.Results of histopathologic changes:The brain tissue of rats in B group were interstitial edema,neuronal cell disorder,cell swelling,light color,smaller nuclei,blurred or even disappear,or visible nerve cell structure was unclear,a large number of microglia hyperplasia,inflammatory cell infiltration;lung tissue can be seen of obvious congestion,alveolar edema,alveolar the interstitial width increased,accompanied by a large number of inflammatory cell infiltration;Obvious necrotic foci were found in the intestinal tissue,which invaded the epithelial layer of the intestinal mucosa,mucosa,submucosa,and necrosis of the intestinal wall.Compared with the B group,the brain edema in the brain tissue of the rats in drug intervention group was reduced,and there were obvious scar like changes in the injured tissue;In lung tissue,there were pulmonary alveolar edema,reduce congestion,alveolar interstitial was widened,with a small amount of powder in the infiltration of inflammatory cells;In intestinal tissue,necrosis and ulcer decreased mucosal epithelium and lamina propria glands arranged neatly,mucosa and submucosa layer to reduce the infiltration of inflammatory cells.4.Results of IL-6 positive expression:Compared with rats in A group,The brain and lung tissues of Rats in B group showed more positive expression of IL-6(P<0.01).The brain,lung and intestine tissues of the positive expression of IL-6 of rats in drug intervention group were lower than B group(P<0.05),the positive expression of E group were at least,compared with the A group had no significant difference(P>0.05).5.The content of VIP and SP:The rats in group B reduce the content of VIP in serum,brain,lung and large intestinal tissues than in group A(P<0.01).In D group and E group rats VIP content in serum,brain,lung and large intestinal tissue is higher than that of B group(P<0.05).Rats in B group of serum,brain and lung tissue with SP content were significantly higher than those in group A(P<0.01),and decreased the content of SP in large intestinal tissue than those in A group(P<0.01).The drug intervention group the content of SP in serum,brain and lung tissues was lower than that of group B(P<0.05),the content of SP in large intestinal tissue is increased than B group(P<0.05)Conclusions:Qing Re Hua Tan Tong Fu Decoction can improve the general state,temperature,weight,defecation and brain nerve function defect in rats with cerebral infarction with bacterial pneumonia.The mechanism may be associated with that the decoction could regulate the content of VIP and SP,down regulated expression of IL-6,to antagonist inflammatory reaction in brain,lung and large intestine tissues.
