| Antrodia camphorata,a rare medicinal fungus mainly distributed in Taiwan.It has intriguing activites like liver protection,anti-tumor and other effects,and it also shows highly medicinal and commercial value.However,due to the scarcity of its wild host resources and the slow growth rate,the large-scale development and application of Antrodia camphorata has be limited.In recent years,the research of the petri-dish cultured Antrodia camphorata has been particularly prominent,and it has become one of the main cultivation methods of Antrodia camphora.Therefore,the petri-dish cultured Antrodia camphorate was chosen as the research object of our study.The separation of triterpenoids in the active fractions and the development of liver-protection and anti-tumor-related activities will be study.The relevant conditions for quality standards will be establishedIn this study,various chromotograhic techniques(silica gel column chromatography,ODS column chromatography and low pressure column chromatography,Sephadex LH-20 chromatography,thin layer chromatography,recrystallization,HPLC etc)had been chose for isolating methanol extract of the petri-dish cultured Antrodia camphorata.Twenty-three compounds were isolated and their structures were identified by nuclear magnetic resonance spectroscopy,mass spectrometry and the physical and chemical properties of the compounds.They included 20 lanolin compounds,one ergosterol type compound and 2 benzene ring type compounds.And 15 of them were the unique composition,only been find in Antrodia camphorata.the peculiarity of these compouds mainly reflected in the substitution of only one methyl group at the C-4 position.In addition,the activity ofthese compounds were assessed by nuclear receptor RXRadual luciferase reporter geneassay,and MTT cytotoxicity.The resultssuggested that coumpounds 6 and 14 showed obvious RXRα transcriptional inhibitory activities.Moreover,compound 12 has the effect on inhibiting the proliferation od HepG-2,but no effect on L02.Moreover,in mice alcohol-induced liver injury model,many index like triglyceride(TG)had been reduce by the petri-dish cultured Antrodia camphorata powder and Antcin K.It shows that the petri-dish cultured Antrodia camphoratacould protect alcoholic liver injury in mice.The quality control of the petri-dish cultured Antrodia camphorata was initially established by using multi-components quantitive by one marker.The HPLC fingerprint conditions of the sample were optimized and the baseline of(25R)-Antcin K and(25 S)-Antcin Kwere separationed firstly.The contents of(25R)-Antcin K and(25S)-Antcin K that difficult to separate were determined by(25S)-Antcin C.At the sane time,the reliability of the results had been proved by external standerd curve.these work laid a solid fundation for quality control of the petri-dish cultured Antrodia camphorata... |
