The Research Of Ergot Steroids From Soild Cultivation Antrodia Camphorata

Antrodia camphorata is a endemic fungi of Taiwan.It has several pharmacological activities such as anti-bacterial,anti-cancer and anti-inflammatory activities.Previous research indicated that ergotsterane steroids were rich in this fungi.However,the chemical study of the solid-stated fermented are Antrodia camphorata scarely reported.For this reason,isolation and structure elucidation of ergotsterane steroids from antrodia camphorata would have great significance for the exploitation and application of antrodia camphorata,In this experiment,solid state cultivated antrodia camphorata was chose as a research object,and solid-stated fermented antrodia camphorata was extracted with 95%ethanol,then the extract was suspended in H2O and separated by silica gel or gel column chromatograph according to the situation of EtOAc soluble fraction in TLC.Consequently,by means of HPLC and TLC,we got a series of Ergotsterane steroids.Eventually,the strcture of these compounds were analyzed by 13C-NMR and 1H-NMR.We reported that Six compounds were identified as following:(3?,22E)-Ergosta-5,7,22-trien-3-ol(1),(22E)-Ergosta-4,6,8(14),22-tetraen-3-one(2),?-sitosterol(3),3?-Hydroxy-5a,8a-epidioxyergosta-6,22-diene(4),Herbarulide(5),Ergosta-7,22-diene-3,6-dione(6),among which compound 1,2,4,5,6 are the ergotsterane compounds and compound 4-6 are isolated from antrodia camphorata for the first time.Literature reports of compound 2 is not only to human laryngeal cancer Hep 2 cells and cervical cancer Hela has cytotoxic activity,and has a strong diuretic.speculation antrodia has certain anti-tumor activity and diuresis,this study laid a solid foundation for further development and utilization of antrodia.
