Antrodia camphorata (A. camphorata), a new basidiomycete distributed only in Taiwan, is well known in Taiwan due to its potent hepatoprotective effect. Due to its rarity in the nature and difficulty in culture, A. camphorata is mainly produced by submerged culture. In this paper, nutritional value, anti-tumor effect of the dry matter of culture broth (DMCB) of A. camphorata in submerged culture were detected in order to evaluate it's exploitation value. The extraction of the main positive compositions and their anti-tumor effects were also preliminary studied.The chemical compositions of A. camphorata in submerged culture and sporoderm-broken spores of G. lucidum were analyzed, including moisture, ash, fat, crude protein, amino acid, triterpenoids, polysaccharides, reducing sugar. The relative content of crude protein, polysaccharides, total phenol, amino acid and reducing sugar were 47.78%, 14.52%, 10.54%, 36.71%, 4.81%, respectively in the DMCB of A. camphorata in submerged culture, which were higher than sporoderm-broken spores of G. lucidum's. Otherwise, the relative contents of fat and triterpenoids in the sporoderm-broken spores powder of G. lucidum were 20.39% and 21.60%, respectively, which was higher than the DMCB'of A. camphorata in submerged culture. Unsaturated fatty acids were prior to saturated fatty acids in the two powders.The extraction of triterpenoids and polysaccharides (fractionⅠand fractionⅡ) were studied. The optimal extraction processing parameters of triterpenoids were as follows: concentration of ethanol 80%, extraction temperature 80℃, the ratio of material to liquid 1:30, extraction time 45 minutes. Under these conditions, the average extracting rate of triterpenoids was 2.60%. The optimal extraction processing parameters of polysaccharides were as follows: extraction temperature 80℃, the ratio of material to water 1:30, extraction time 2 hours, extraction times 2 times. Under these conditions, the average extracting rate of polysaccharides was 20.02%.To investigate the inhibitory of the samples on the growth of mouse cancer (H22), a model of the solid tumor of transplanted cancer (H22) were built. The rate of the inhibition of tumor growth, the indexes of spleen and thymus, the time of survival, MDA,SOD,GSH-Px of mice liver were checked. The pathological change of solid tumor and the expression of bax/bcl-2 gene albumen in tumor tissues were also observed. The results suggested that the DMCB of A. camphorata in submerged culture and its main positive compositions (fractionⅠand fractionⅡ) inhibited the growth of tumor, prolonged the survival time of mice, reduced bcl-2 protein expression, improved bax protein expression, increased anti-oxidative enzymatic activity in mice liver homogenate in different degree and had no toxic effect to immune organs. The polysaccharides (fractionⅡ) showed the best effect in inhibiting the growth of tumor in vivo. The DMCB of A. camphorata in submerged culture was as good as the sporoderm-broken spores of G. lucidum on anti-tumor effect.
