Experimental purposes:The causes of peptic ulcers are complex,and the emotional distress plays an important role.Modified Xiaojianzhong Decoction is an effective prescription for treatment of peptic ulcer that is through years of clinical study by prefessor Qian Linchao.The effect of Modified Xiaojianzhong Decoction on Brain-gut Axis pathway in the treatment of peptic ulcer is the focus of the study.This study observed the behavioral characteristics,weight change,ulcer index and morphological changes of gastric mucosa in each experiment,and selected the NTS,NPY,PYY,and GHRL of BGP in close relationship with emotional factors and mentalstimulation for the study of indicators to detect changes in the level of their content.This paper explored the repair effect of gastric ulcer and the regulation of Brain-gut Axis,and it also provided the effect basis for clinical medication.Experimental Method:32 Healthy and Clean Level SD rats,10-12 weeks,half male and female,weight(200 ± 20 g),after adapting to the environment,randomly divided into 4 groups:There are Normal group,Model group,Western medicine group,Chinese medicine group,each group with 8 rats.With restraint cold water immersion stress(WRS)method,resulting in a rat model of gastric ulcer.Rats in the Normal group were not modeled.Model group,Western medicine group and Chinese medicine group modeled by WRS gastric ulcer were treated respectively by the normal saline,Omeprazole western medicine water suspension,Modified Xiaojianzhong Decoction,and treatment of 14 days.The experimental steps are as follows:(1)measuring and comparing the weight changes before and after the treatment of each group;(2)using the HE staining method to observe the gastric mucosal morphology;(3)comparing ulcer index and ulcer inhibition rate;(4)using enzyme-linked immunosorbent assay(ELISA)detect the content level change of serum,gastric tissue,and cerebral cortex of each group of rats Neurotensin(NTS),Neuropeptide Y(NPY),Peptide tyrosine tyrosine(PYY),Ghrelin(GHRL)and other important indexes.Experimental results:1.Weight of rats:Compared with model group,the body weight of Western Medicine Group and Chinese Medicine Group increased significantLy(p<0.01 or p<0.05);Compared with Chinese Medicine Group,Chinese Medicine Group had higher body weight than Western Medicine Group(p<0.05).2.Rat ulcer index:Compared with model group,both Western Medicine Group and Chinese Medicine Group can reduce ulcer area(p<0.01).Chinese Medicine Group is significantly superior to the Western Medicine Group(p<0.05).Compared with model group,the ulcer inhibition rate of Western Medicine Group was 56.66%,and the ulcer inhibition rate of Chinese Medicine Group was 76.66%,and Chinese Medicine Group was significantly superior to Western Medicine Group(p<0.05).3.Effect of rat NTS content level:compared with model group,in serum,stomach,and cerebral cortex of the detection,the content of Western Medicine Group and Chinese Medicine Group significantly reduced(p<0.01 or p<0.05).In the cerebral cortex aspect,comparing with Western Medicine Group,Chinese Medicine Group's content level is closer to Normal Group(p<0.05).4.Effect of levels of rat NPY concentration:In serum,stomach,cerebral cortex of the test,content of Western Medicine Group and Chinese Medicine Group significantly reduced(p<0.05),and the levels of the two groups are close to Normal group.5.Effect of levels of rat PYY concentration:in serum detection compared with model group,the content of Western Medicine Group and Chinese Medicine Group decreased significantly(p<0.05),and the levels of the two groups are close to the Normal Group.Compared with the model group,the contents of Western Medicine Group in the stomach and cerebral cortex were significantly reduced(p<0.05),which is closer to the normal group.However,there was no significant difference in the content of Chinese Medicine Group(P>0.05);Compared with Chinese Medicine Group,Western Medicine Group's content level is closer to Normal Group(p<0.05).6.Effect on content of rat GHRL:In serum,in the stomach,in the cerebral cortex,the contents of Western Medicine Group and Chinese Medicine Group were significantly reduced(p<0.01 or<0.05).In the serum,the Western Medicine Group and the Chinese Medicine Group,the Chinese Medicine Group's content level is closer to the normal group(p<0.05).Experimental conclusion:Through the experiment,it is concluded that Modified XiaojianzhongDecoction can effectively repair and reduce gastric tissue mucosal injury in rats with gastric ulcer,and the effect of Modified Xiaojianzhong Decoction on the healing of ulcer in experimental rats is significant.The effect of Modified Xiaojianzhong Decoction on the cure of PU is to reduce the NTS,NPY,PYY,and GHRL of BGP in the serum,stomach,and cerebral cortex to heal the ulcer,which is influenced by Brain-gut Axis.The effect of Modified Xiaojianzhong Decoction can mobilize the level of BGP and stimulate the balance of gastric mucosal protective factors and weakening gastric mucosal attack factors,so that the PU treatment has a positive effect.However,a further experimental study is needed to demonstrate that if there is a specific quantitative relationship between the optimal physical level threshold and the BGP for each BGP in healthy state.
