Based On The Brain-gut Interaction And The Effect Of Wei Liqi Recipe On Brain-gut Peptides In Rats With Functional Dyspepsia

Objective:Through observation and Heweiliqi parties to change the type of functional dyspepsia rats gastric emptying function and the hypothalamus and Ghrelin protein expression in the stomach,the influence of content of Nesfatin-1 in plasma.From the aspects of brain gut interactive type for liver and Heweiliqi party,possible mechanisms of functional dyspepsia.Materials and methods:Will only 40 healthy female Wistar rats,according to the random number table method in rats were randomly divided into 2 groups,the normal group(10)and model group(30).After 7 days of adaptive feeding,Model group on irregular feeding and clip tail stimulation double factor method change the type of functional dyspepsia rats model was established,Blank group of conventional breeding,A total of 21 days.Closely observed during rat rat's general status.When the molding was done,The rats were randomly divided into model group and Heweiliqi group,the Mautolin group and model control group,each group of 10.Rats in the Heweiliqi Formula group were gavaged with 0.9702g/ m L of Heweiliqi Formula solution,and rats in the Mautoline group were gavaged with 0.25mg/ m L of Mautoline suspension.Blank group and model group during the period of drug group was given drug treatment,at the same time take such as volume of 0.9% saline water irrigation.Twice daily for 14 consecutive days.After the treatment,observation records each rat's general status and comparing with before.At the end of the study,gastric emptying rates were measured for all rats to assess gastrointestinal motility,Determination of rat hypothalamus and protein expression of Ghrelin in stomach tissue and serum Nesfatin-1 content.Result:1.Compared with the blank group,the rats in the model group were easily frightened,irritable,dull hair,slow reaction,anorexia and water dislike,and the gastric empty-rate was significantly reduced(P<0.05),indicating the success of modeling.2.Compared with the blank group,the expression levels of Ghrelin in gastric antrum and hypothalamus of FD rats with liver depression were significantly decreased(P<0.05,P<0.05),and the expression levels of Ghrelin protein in gastric antrum and hypothalamus of rats in He Wei Li Qi Formula group and Morbutoline group were significantly increased(P<0.05,P<0.05).The expression of Ghrelin protein in the stomach of rats in the morbutoline group was slightly lower(P<0.05).3.Compared with the blank group,serum Nesfatin-1 content of rats in liver depression type FD model group was significantly increased(P<0.05),and after drug treatment,serum Nesfatin-1 content of rats in He Weiliqi prescription group and Morbutoline group was significantly decreased(P<0.05,P<0.05).Conclusion:1.The experiment successful replication type liver depression FD rats model,By comparing the general condition of rats before and after the treatment,The results showed that Heweiliqi formula could significantly improve the mental state of functional dyspepsia rats with liver depression.2.By comparing the gastric emptying rate of rats in each group,The gastric emptying rate in Heweiliqi formula group and Morbutoline group was significantly higher than that in model group.The results showed that He Wei Li Qi Formula and Morbutoline could better promote gastrointestinal motility.To improve gastrointestinal symptoms of functional dyspepsia rats.3.By examining each rat hypothalamus and the expression of Ghrelin in stomach tissue protein,It was found that compared with the blank control group,the expression of Ghrelin protein in hypothalamus and stomach tissue of rats in the model group was significantly decreased.Heweiliqi decoction up-regulated the expression of Ghrelin protein in hypothalamus and stomach tissue of FD rats with liver depression.It is speculated that Heweiliqi formula may improve gastrointestinal motility and depression state of FD rats with liver depression by participating in regulating brain-gut interaction.4.By detecting the serum Nesfatin-1 content of rats in each group,Compared with the blank control group,the serum Nesfatin-1 content of rats in model group increased.And He Wei Li Qi Formula can significantly reduce the serum Nesfatin-1 content of FD rats with liver depression.It was suggested that Heweiliqi Formula may promote appetite and feeding behavior of FD rats with liver depression by down-regulating the expression of Nesfatin-1.It is speculated that Heweiliqi formula can improve the depression and appetite of FD rats with liver depression by participating in the regulation of brain-intestinal peptide.It has good advantages to treat functional dyspepsia from many aspects.
