To Explore The Therapeutic Of Sgp130 On Rheumatoid Arthitis

Objective:The soluble gp130(soluble gp130,sgp130)protein was used as a drug for rheumatoid arthritis.The therapeutic effect of sgp130 on rheumatoid arthritis was explored by injecting sgp130 into the tail vein of mice by successfully constructing mouse rheumatoid arthritis model.Methods:We established the Prokaryotic expression system:pET28a-sgpl30,expressed and purified the protein of sgp130.Using the bovine type II collagen to induce arthritis in DBA/1J mice,and the different doses of sgpl30 protein were injected into tail vein.The therapeutic effect of sgpl30 on rheumatoid arthritis was studied as follows:the degree of swelling of the joints was measured from the day of sensitization,the levels of rheumatoid factor and related inflammatory factors,and the liver function indictors were measured by blood test.Immunohistochemical method was used to detect the degree of internal organ damage such as paw joints and liver.Results:1.We successfully established the bovine type ? collagen-iduced(CIA)is an animal model of rheumatoid arthritis.2.Sgp130 can effectively improve the clinical score of CIA mice and paw swelling and other general scores,indicating that it has a certain therapeutic effect.3.Sgp130 protein can significantly reduce ser?m levels of IL-6,TNF-a and other inflammatory cytokines.4.Blood testing show that sgp130 protein can reduce liver function indicators significantly,such as:ALT.5.Paw joints were obtained at the end of the experiment,joint tissues were stained with hematoxylin-eosin stains and analyzed.Paw joints of CIA-mice demonstrated extensive inflammatory cell infiltration,synovial hyperplasia,cartilage and bone erosion.Sgpl30 recipients developed a less cellular infiltration with smooth articular surface and wider articular space,lesser cartilage erosion or bone damage which is comparable to that of naive,and better than MTX groups.6.The livers of mice were stained with hematoxylin-eosin stains showed that sgp 130 recipients had homogeneous liver texture,smooth capsule and slender liver,and the degree of liver fibrosis was significantly decreased.7.From the objects of lung and the lung tissues were stained with hematoxylin-eosin stains that show,sgp130 can protect lung from damagement.Conclusion:We found that sgp130 effectively reduced the serum levels of TNF-a,IL-1?,and IL-6;maintained the bone surface pattern;and decreased both paw swelling and the development of gross lesions,in the CIA mice model.These results suggest that sgp130 may be useful to treat both RA and other inflammatory disorders.Compared with the model group,the kidney,liver,and other organ damage was significantly reduced.This study can be concluded that sgpl30 on rheumatoid arthritis does have a certain effect,and its side effects relative to MTX smaller,these results indicate that sgp130 can be used to treat RA and other inflammatory diseases,for the future clinical treatment of rheumatoid Arthritis has a good research significance.
