Cancer has become the second leading cause of death in the world.Breast cancer is one of the most common malignant tumors in women,which seriously threatens women's physical and mental health and life.At present,the traditional treatment for breast cancer is surgery,radiotherapy and chemotherapy.These treatments have great side effects and destroy the immune regulation ability of the body,and also bring great pain to the patients.Therefore,it is very important to find the substances that have anti breast cancer activity.It is reported that Sparasiscrispa polysaccharide has a variety of bioactivity and has good antitumor activity.However,there is little research on its activity against breast cancer.Therefore,we evaluated the anti breast cancer activity of Sparasiscrispa polysaccharide by two aspects in vivo and in vitro.This study mainly includes the following two aspects:1.Study on anti-breast cancer activity of Sparasiscrispa polysaccharide and its mechanism in vitro.In this paper,we have shown that the growth and migration of MCF-7 and MDA-MB-231 cells can be inhibited by the Sparasiscrispa polysaccharide through MTT test,LDH detection and Migration assay.The mechanism of the anti breast cancer activity of Sparasiscrispa polysaccharide was investigated by biochemical analysis.It was found that Sparasiscrispa polysaccharide can induce apoptosis of mammary gland,and increase the level of ROS,decrease mitochondrial membrane potential,activate caspase cascade,increase the expression of Pro apoptotic protein,and decrease the expression of inhibitor of apoptosis protein.In conclusion,in vitro Sparasiscrispa polysaccharide can induce apoptosis of breast cancer cells through mitochondrial pathway,and inhibit the migration and growth of breast cancer cells.2.Study on anti-breast cancer activity of Sparasiscrispa polysaccharide and its mechanism in vivoIn order to further determine the anti breast effect of Sparasiscrispa polysaccharide,an ectopic tumor model of MCF-7 nude mice was established.After 14 days of treatment,the tumor volume decreased significantly,and the weight and pathological sections of nude mice showed no significant change compared with the blank group.The expression of mitochondrial apoptosis related protein was detected by Western blot and the results were in agreement with the experimental results in vitro.At the same time,we also found that the spleen and thymus index of the tumor bearing mice increased after 14 days after the therapy.And the content of cytokines associated with immunization also increased.Therefore,we can conclude that Sparasiscrispa polysaccharide can not only inhibit tumor growth by inducing apoptosis,but also play an anti-tumor activity by enhancing the body's immune function.To sum up,Sparasiscrispa polysaccharide has anti breast cancer activity,it can not only induce apoptosis of breast cancer cells through mitochondrial apoptotic pathway,but also act as an immunomodulator,induce the secretion of immune cytokines and achieve the inhibition and killing of tumor cells.
