This paper studies the effect of inhibiting the proliferation of human astragalus selenium polysaccharide on cultured breast cancer MCF-7 tumor cells and to explore the mitochondrial membrane potential,reactive oxygen species in the cells,the cells in calcium ion concentration of selenium with astragalus polysaccharides induce breast cancer cells relationship apoptosis.In the in vitro study,the astragalus selenium polysaccharide can inhibit the growth MCF-7 cells.Astragalus measured by MTT assay in vitro selenium polysaccharide on MCF-7 cells,the IC5o is 14.65?g/ml,and the experimental results confirm this inhibition was a significant dose-dependent manner.experimental results show that selenium astragalus polysaccharides on human breast cancer cell growth inhibition play to achieve anti-cancer effects.Thus the experimental results of tlhe study by the above two methods show that selenium astragalus polysaccharides on growth MCF-7 cells play a significant inhibition.Through further study on cell morphology confirmed the role of selenium astragalus polysaccharides on human breast cancer cells induce apoptosis.Astragalus Polysaccharide different concentrations of selenium in MCF-7 cells for 48 hours,by observing the group can see the state of cells in the negative control group and arranged very densely,deformed or broken cell does not exist,and as the administration increasing concentration,intensity gradually becomes sparse cell growth,cell shape shrinkage,high concentrations in most cells broken,broken cell morphology,without a complete profile,the number of adherent cells decreased.These observations confirmed that selenium Astragalus polysaccharides on MCF-7 cells apoptosis,and it was a significant dose-dependent.Next,using fluorescence microscopy on cell morphology more detailed observation,astragalus polysaccharide different concentrations of selenium in MCF-7 cells after 48h,using Hoechst33258 fluorescent dye staining of human breast cancer cells,and by observing the fluorescence microscope and found that Astragalus with increasing doses of selenium polysaccharide clearly observable phenomena characteristic of apoptosis,negative control group showed MCF-7 cells were uniform chromatin,nuclear morphological rules,nucleus fragmentation dose group,high dose group nuclei it was dense stain,which indicates that astragalus polysaccharide selenium can lead to the appearance of morphological changes in human breast cancer cells.Using flow cytometry analysis showed that different concentrations of selenium astragalus polysaccharides MCF-7 human breast cancer cells cells after 48 h,with a clear peak of apoptosis and apoptosis rate astragalus selenium polysaccharide group was(0.47±0.41)%,(13.89±2.93)%,(33.04±2.08)%,and the peak of apoptosis significantly with the administration of selenium concentration Astragalus polysaccharides showing a significant dose-dependent.Further illustrates the astragalus selenium polysaccharide on human breast cancer MCF-7 cells from apoptosis induced by quantitatively.Finally,according to the above experiment,based on the mechanism of induction of Astragalus selenium polysaccharide MCF-7 human breast cancer were studied,mainly using laser scanning confocal microscope astragalus polysaccharides on human selenium MCF-7 breast cancer cells for 48 hours,the cells active changes in oxygen,changes in calcium concentration in cells and changes in mitochondrial membrane potential of the cells were observed.Mitochondrial membrane potential was observed by fluorescence intensity was 32.47±1.45,23.49±0.58?20.50±0.88(membrane potential of the negative control group was 38.05±2.30).Reactive oxygen fluorescence were:negative contro26.98 ±0.43,52.15±0.61,79.00± 1.06.Calcium fluorescence intensity were:negative control 5.103 ±0.14,23.41±0.53,47.37±0.92 ? 75.90±1.14.Each group were statistically significant(P<0.05)compared to the control group.Through the above results show that selenium astragalus polysaccharides on human breast cancer cells was significantly inhibited,but also through an inverted microscope,fluorescence microscope and observed by flow cytometry on a single PI staining for apoptosis rate of adoption,regardless from a qualitative and quantitative point of view perspective,confirms the role of co-Astragalus selenium polysaccharide on human breast cancer cells MCF-7 can induce apoptosis.And by studying the results of the mechanism of action suggests that the effects of Astragalus polysaccharide selenium into the cell,with the administration of increasing concentrations of mitochondrial membrane integrity is destroyed,and showing changes in membrane potential,reactive oxygen content increased,the calcium ion concentration swells,so that the mitochondrial membrane channel pore opening,leading to apoptosis.This suggests that selenium astragalus polysaccharides on the human body through the mitochondrial pathway MCF-7 breast cancer cell apoptosis.In summary,in vitro studies confirmed that selenium astragalus polysaccharides can induce human breast cancer MCF-7 apoptosis pathway and apoptosis is likely to be the mitochondrial pathway.
