The experimental study of Capparis spinosa selenium polysaccharide on human bre ast cancer MCF-7 cell growth inhibition and apoptosis,and discuss its pathways and m echanisms.Capparis spinosa selenium polysaccharide detect cytotoxicity of human breast cance r MCF-7 cells by MTT assay in this experiment.We observe that Capparis spinosa sele nium polysaccharide induce morphological changes of apoptosis of MCF-7 cells by inve rted microscope and fluorescence microscopy.We detect different concentrations of seleni um polysaccharide Capparis spinosa on apoptosis rate of MCF-7 cell by flow cytometry PI staining.we detect Capparis spinosa selenium polysaccharide to MCF-7 cells about th e changes of Ca2+ concentration,mitochondrial membrane potential,reactive oxygen specie s(ROS)by confocal laser scanning microscopy(CLSM).Experimental results show that MTT assay showed that Capparis spinosa selenium polysaccharide on human breast cancer MCF-7 cells with cytotoxic effect.Capparis spino sa selenium polysaccharide effect human breast cancer MCF-7 cells after 48h,we obser ve adherent growth of negative control MCF-7 cells under an inverted microscope,clos ely arranged,diamond-shaped cell and integrity of cell membrane.The concentration of of positive control group is amycin.Wrinkling cell disruption of the most cell surface,cell morphology is incomplete.Capparis spinosa seleniumpolysaccharide groups increase administration dosage and cell density change sparse,loose,smaller size,phenomena of wrinkling deformation,etc.We observe MCF-7 cell morphology by Hoechst33258 fluor escent staining.Cell chromatin of negative control MCF-7 cells is still uniformity intacti ng no rupture wrinkling.The concentration of Capparis spinosa selenium polysaccharide increase in each group,meantime with the increase of the number of apoptotic bodies,t he density of cell growth gradually becomes sparse,wrinkled surface,partially broken ce lls,cell morphology become incomplete.The influence of Capparis spinosa selenium polysaccharide on MCF-7 cell apoptosi s detect by flow cytometry and PI staining.Capparis spinosa selenium polysaccharideeffe ct human breast cancer MCF-7 cells after 48h,tumor cell appear the peak of apoptosis significantly,with the increasing dose of tumor cells also increased the number of apop tosis.Description that inhibition of Capparis spinosa selenium polysaccharide to MCF-7 i s related to inducing apoptosis of tumor cell.Capparis spinosa selenium polysaccharide o n Ca2+ concentration of MCF-7 cells detectd by confocal laser scanning microscopy.The experimental results,the higher dose increases the Capparis spinosa selenium polysaccha ride,the taller increase of Ca2+ concentration.The changes of membrane potential showe d that with increasing of dose concentration decreased the value of membrane potential.The study of reactive oxygen species(ROS)showed that with increasing doses of Cappa ris spinosa selenium polysaccharide,ROS elevated the levels intracellular,and the contr ol group were significantly different.In summary,Capparis spinosa selenium polysaccharide can inhibit the proliferation of human breast cancer MCF-7 cells,and the ability to induce apoptosis in MCF-7 cells.
