The experimental study of Sargassum fusiforme seleno-polysaccharide on human breast cancer MCF-7 cell growth inhibition and apoptosis,and discusses its pathways and mechanisms.MTT showed that Sargassum fusiforme seleno-polysaccharide on human breast cancer MCF-7 cells have a certain inhibitory effect with increasing concentrations,and with the Sargassum fusiforme seleno-polysaccharide concentration increases inhibition enhances,the human breast cancer MCF-7 cells IC50 7.94?g/ml.Through the inverted microscope observation in direct Sargassum fusiforme seleno-polysaccharide human breast cancer MCF-7 cells 48 h,negative adherent cell intact,clean cut,without cell fragmentation and wrinkling.With the concentration increased cell growth density gradually thinning,cell shrinkage,high concentration group most of cell disruption,cell morphogenesis is complete,adherent to reduce.Description of Sargassum fusiforme seleno-polysaccharide on human breast cancer MCF-7 cell are apoptosis,and in a dose-dependent.Observed under fluorescence microscopy after Hoechst33258 staining,normal cells showed uniform blue fluorescent nuclei,and apoptotic cells nucleus showed a dense or chunky concentrated dye fluorescence.The experimental results show that,Sargassum fusiforme seleno-polysaccharide role for human breast canner MCF-7 cells after 48 h,the negative control group the results showed that human breast cancer MCF-7 cells were uniform chromatin,nuclear morphology rules,in dosage group nuclei fragmentation,high dosage group of nuclei showed a dense,this suggests that Sargassum fusiforme seleno-polysaccharide can result in MCF-7 cell morphological change,and the cells change with the increase of dosage,appeared phenomenon of apoptosis.The experimental use of propidium iodide single staining by flow cytometry analysis showed that different concentrations of Sargassum fusiforme seleno-polysaccharide role of human breast cancer MCF-7 cells after 48 h,have a obvious apoptosis peak,each cell apoptosis rates were(2.85±0.08)%,(24.36±3.89)%,(31.58±2.72)%,as the drug concentration increased,further description of Sargassum fusiforme seleno-polysaccharide inhibition effect on human breast cancer MCF-7 cells and inducing tumor cell apoptosis related.On the basis of these experiments,the experiment also investigated the Sargassum fusiforme seleno-polysaccharide induces apoptosis in human breast cancer MCF-7 cells apoptosis.Using confocal laser scanning microscopy to detect changes in mitochondrial membrane potential of human breast cancer MCF-7cells different concentrations of Sargassum fusiforme seleno-polysaccharide role in human breast cancer MCF-7cells after 48 h,mitochondrial membrane potential of fluorescence intensity were:low-dose 44.09±0.14,dose 25.64±0.25,high-dose 3.82±0.09.Using confocal laser scanning microscopy to detect changes in calcium levels of human breast cancer MCF-7cells,different concentrations of Sargassum fusiforme seleno-polysaccharide role in human breast cancer MCF-7 cells after 48 h,calcium of fluorescence intensity were:low-dose 8.36±0.41,dose 11.86±0.13,high-dose 34.13±0.94.Using confocal laser scanning microscopy to detect changes in reactive oxygen species of human breast cancer MCF-7cells,different concentrations of Sargassum fusiforme seleno-polysaccharide role in human breast cancer MCF-7cells after 48 h,reactive oxygen species of fluorescence intensity were:low-dose 34.30±0.34,dose 61.60±0.72,high-dose 154.36±3.72.These results show that Sargassum fusiforme seleno-polysaccharide induce human breast cancer MCF-7 cells apoptosis through mitochondrial pathway:Sargassum fusiforme seleno-polysaccharide effects to cells after administration,as increasing dose as the mitochondrial membrane is damaged,the mitochondrial membrane permeability increased,so that the mitochondrial membrane potential,reactive oxygen species increased,calcium ion concentration,leading to mitochondrial membrane channel pore opening unduly,and the cells apoptosis finally.In summary,this thesis demonstrated Sargassum fusiforme seleno-polysaccharide can induce human breast cancer MCF-7 cell apoptosis by in vitro experiments and mitochondrial pathway is Sargassum fusiforme seleno-polysaccharide induces apoptosis in human breast cancer MCF-7 cell apoptosis may be one way.
