Association Of RAC2 Gene Polymorphism With Susceptibility Of Chronic Periodontitis Among Chinese Northwestern Population

Objective: Periodontal tissues of patients with chronic periodontitis of long-standing chronic inflammation can damage periodontal tissue.Studies have shown that oral health is not just a problem of chronic periodontitis,but also with diabetes,cardiovascular diseases,gastrointestinal diseases and other systemic diseases are associated.With the development of molecular biology technology and platform,the methods of Transcriptomics and Proteomics have been a reliable means to study the molecular mechanism of disease and to screen biomarkers.This paper discusses the differential expression genes and differentially expressed proteins in gingival tissues of healthy people and chronic periodontitis patients,and further explores the association between the differentially expressed gene RAC2 gene polymorphisms and the risk of chronic periodontitis.Methods: In this paper,the differential expression genes between the two groups were analyzed by RNA sequencing data(GSM2133915 and GSM2133916)in the healthy population and chronic periodontitis patients provided by the Gene Exression Omnibus Database and the differential expression genes involved in biological process and cell signaling pathway analyzed by DAVID enrichment tools.The gingival tissue samples of 21 patients with chronic periodontitis and 21 healthy control subjects were collected in this study,and the differential expression proteins between the two group tissue samples was analyzed by using two-dimensional gel electrophoresis combined with mass spectrometry.The peripheral blood samples of 231 patients with chronic periodontitis and 195 healthy control subjects were collected in this study,using polymerase chain reaction-restriction fragment length polymorphism analysis the rs6572 and rs1476002 genotype of RAC2 gene after extracting genomic DNA of the samples in each group and verified by sequencing.The genotyping data were analyzed by SPSS19.0 software.SHEsis software for linkage disequilibrium analysis.Results: 1.In this study,we found that there were 1184 differentially expressed genes in GSM2133915 and GSM2133916,among which 415 were low expression genes and 769 were high expression genes in chronic periodontitis group.DAVID enrichment revealed that the differential gene was mainly associated with biological processes such as immune response,inflammatory response,cell adhesion,apoptosis,proteolysis,stress response,bone system development and other biological processes.2.Furthermore,the protein expression of gingival tissue in healthy people and chronic periodontitis patients was compared by proteomics research method.It was found that serum albumin,which was conducive to repair the injured tissue,was relatively low in the gingival tissues of patients with chronic periodontitis.Fibrinogen,Annexin A2 associated with inflammatory response,Ig G associated with immune response,and triosephosphate isomerase involved in glycolysis expression level was relatively high in gingival tissue of patients with chronic periodontitis high.3.Based on the distribution of rs6572 and rs1476002 loci of RAC2 gene and alleles in healthy and chronic periodontitis patients,it was found that the rs6572 and rs1476002 were found in the distribution of genotypes and alleles with chronic periodontitis susceptibility had no correlation.The analyses were stratified by gender and found that the rs6572 GG genotype was found to be associated with the risk of chronic periodontitis in women(P=0.043,OR=2.554,95%CI=1.023-6.374),rs6572 G allele was a risk factor for chronic periodontitis in women,and the risk of chronic periodontitis was increased by 1.565 times(P=0.046,OR=1.565,95%CI=1.007-2.433).Conclusion: In this study,we analyzed the differential expression genes and proteins of gingival tissue in chronic periodontitis patients and healthy people,and preliminary found molecular pathogenesis and potential biomarkers of chronic periodontitis.The correlation analysis between RAC2 gene polymorphisms and susceptibility to chronic periodontitis suggest that there were no significant correlations between rs6572 and rs1476002 and the risk of chronic periodontitis,but the rs6572 polymorphisms and risk of chronic periodontitis with gender differences.This work provides a certain amount of data for screening biomarkers of chronic periodontitis.
