Association Between STAT4 Gene Polymorphism And Diabetes With Chronic Periodontitis Susceptibility In A Chinese Population

Background:Type 2 diabetes mellitus and chronic periodontitis are caused by chronic and complex disease, which are the result of combined action of genetic and environmental factors. Periodontitis was described as the sixth complication of diabetes. Signal transducer and activator of transcription 4 (STAT4) can induce inflammatory response. This study aims to investigate the association between STAT4 genetic polymorphisms and type 2 diabetes(T2DM)、chronic periodontitis(CP)、type 2 diabetes with chronic periodontitis (T2DM&CP) in a Chinese population.Method:A case-control association study was conducted, the samples were divided into four groups:CP (n=255)、T2DM (n=235)、T2DM&CP (n=215) and healthy control (n=262). Three SNPs (rs3821236, rs10168266,rs16833260) polymorphism of the STAT4 gene were tested by combining polymerase chain reaction-restriction fragment length polymorphism method (PCR-RFLP) with direct sequencing method. The obtained result was analyzed by chi-square test of SPSS 19.0. And it was analyzed the types of the haplotypes reconstructed by using the SHEsis software.Result:Compared to case-control groups, the AA genotype of rs3821236 was a protective factor of T2DM、CP and T2DM&CP [P=0.002,OR=0.430; P=0.002, OR=0.451; P=0.001, OR=0.448]. The TT genotype of rs10168266 was associated with increased risk of T2DM and T2DM&CP [P=0.007, OR=1.983; P=0.002, OR=2.249].The GG genotype of rs16833260 was a risk factor of T2DM and T2DM&CP [P=6.75e-005,OR=2.630;P=8.07e-005,OR=2.790].The ATC appeared to be a protective haplotype for CP、T2DM and T2DM&CP[P=0.0013,OR=0.527; P=1.77e-010,OR=0.179; P=3.19e-013,OR=0.079].Conclusion:The results showed that three SNPs polymorphism of STAT4 (rs3821236、 rs10168266、rs16833260) gene are significantly associated with T2DM and T2DM&CP in a Chinese population. Moreover, it was found that the polymorphism of rs3821236 was also associated with the susceptibility of CP in a Chinese population.
