Objective: To investigate the association between single nucleotide polymorphisms(SNP)and chronic pancreatitis(CP)in PRSS1 gene.Methods: DNA samples were collected from patients with chronic pancreatitis and healthy controls.DNA was extracted.5 SNPs in the PRSS1(rs2011216,rs10273639,rs10246334,rs2367343,rs9969188)were selected and genotyped by Snapshot methed.All the statistical analyses were conducted by SPSS software.At the same time,the biochemical indexes of chronic pancreatitis patients and healthy controls were collected,Whether long-term drinking,smoking,had biliary disease and so on.Clinical manifestations and complications of chronic pancreatitis and Imaging performance(Including pancreatic atrophy,pancreatic calcification,pancreatic duct stones,pancreatic duct dilatation)were collected(Including abdominal pain,radiation pain,weight loss,diabetes mellitus,pancreas,pancreas,pancreas,pancreas).to explore the correlation between PRSS1 gene single nucleotide polymorphisms and CP clinical manifestations and complications,imaging performance.Results: There were 89 patients with chronic pancreatitis,192 healthy controls.The number of long-term drinking,smoking,suffering from biliary tract disease in Chronic pancreatitis group was more than in control group,the difference was statistically significant(P<0.05).The P value of the Hardy-Weinberg equilibrium test was higher than 0.05 for the five SNP loci in the case group and the control group.The frequency of the minimum allele G of the single nucleotide polymorphism locus rs2011216 was higher in the case group than in the normal group(P <0.05).Individuals with G at the rs2011216 site had chronic pancreatitis.The risk will rise(OR = 1.66,95% CI = 1.14-2.41).There was no significant difference in the allele distribution of rs10273639,rs10246334,rs2367343 and rs9969188 single nucleotide polymorphisms in CP group and control group.The rs2011216 locus on the PRSS1 gene was associated with an increased risk of chronic pancreatitis in individuals with rs2011216GG(P = 0.018,OR = 2.48,95% CI = 1.15)in the homozygous model compared with the rs2011216 AA population(P = 0.025,OR = 1.79,95% CI = 1.07-2.98).In the dominant model,the risk of chronic pancreatitis was increased in individuals with rs2011216 AG + GG compared with rs2011216 AA).There were 32 cases of smoking in 89 patients with CP.In the rs9969188 locus dominant model,the frequencies of AA genotype in 32 patients with CP were higher than those in the normal group(P <0.05).The OR value was 2.19,95% CI 1.01-4.73.For the same SNP locus,the difference in the same clinical manifestations between the three different genotypes was not statistically significant(P> 0.05).Conclusions: 1.The risk of chronic pancreatitis in individuals with risk allele G on rs2011216 locus with PRSS1 gene was increased.2.In the rs9969188 locus dominant model of smokers,the rs9969188 locus GG The risk of chronic pancreatitis in individuals with rs9969188 AA was increased.3.The polymorphism of PRSS1 gene levels was not correlated with phenotype and there was no causal relationship.
