A dry and mature seeds of Celosia argentea L.is Celosiae semen,a species of Amaranthaceae,which is loaded into a pharmacopoeia.Celosiae semen is bitter,slightly cold,and mainly pass through liver channel.Its main function is to retreat cataract,and clear liver fire,so it is used to cure eyes disease,which is in the recorded"Shen Nong's herbal classic","Compendium of Materia Medica"and other ancient books.Celosia argentea L.is distributed in a lot of parts of China,and also widely survived in other tropical and subtropical countries.Modern pharmacological studies have shown that seeds of Celosia argentea L.has functions of anti-tumor,liver-protecting,blood-sugar-reducing,cataracts treatment.The main chemical compositions are saponins,cyclic peptides,alkaloids,amino acids,protein,minerals and so on.This paper systematically studied the chemical constituents of the 50%ethanol extract of Celosiae semen were 12 compounds were isolated by using macroporous resin column chromatography,silica gel column chromatography,ODS,MCI and HPLC.12 compounds were identified by spectral data(1H-NMR,13C-NMR,HMBC,HMQC,ESI-MS and UV).They were CelosinI?compound 1?;?-spinasteryl-3-O-?-D-glucoside?compound 2?;CelosinJ?compound 3?;CelosinH?compound 4?;2,23-dyhydroxy-3-O-[?-D-xylopyranosyl-?1?3?-?-D-glucuronopyranosyl]–28-O-?-D-glucopyranosyl-olea nolic acid?compound 5?;CelosinK?compound 6?;Chikusetsusaponin??compound 7?;2?-hydroxy-23-aldehyde-3?-O-[?-D-xylopyranosyl-?1?3?-?-D-glucuronopyranosyl methyl ester]oleanolic acid?compound 8?;benzyl cyanide-4-O-?-D-apiofuranosy-?1?6?-O-?-D-glucopyranoside?compound 9,osmanthusideH?;benzyl cyanide-4-O-?-L-rhamnopyranosyl-?1?6?-O-?-D-glucopyranoside?compound10?,daucosterol?compound 11?,CelosinA?compound 12?,of which compound 10 and compound 12 are the new compounds.And compound 2,compound 5,compound 7 and compound 9 were isolated from Celosia argentea L.for the first time.HPLC-UV methods was established from oleanolic saponins and nitrile components in seeds of Celosia argentea L.,and 12 batches of seeds from different soures were examined the effective component in this way.This experiment examined the contents of 5 oleanolic acid saponins and two nitrile compounds,and compared the contents of 12 batches of Celosiae semen,.The results showed that there were significant differences in the contents of seven compounds from 12 batches of Celosiae semen.The antibacterial activity of eight different parts?Fr.1-Fr.3 and Fr.2-1-Fr.2-5?was evaluated by the disc diffusion method,and the antibacterial activity site Fr.2-3 was separated to obtain three the compounds:CelosinI,CelosinJ,and CelosinH evaluation antibacterial activity.These results show that:Fr.2-3 had obvious bacteriostatic effect on Monilia albican and Saccharomyces cerevisiae with MIC of 25mg/mL and 6.25 mg/mL,and MIC of celosinJ and celosinH were 2.5mg/mL and 2.7mg/mL on Saccharomyces cerevisiae.At present,the incidence of AD?Alzheimer's disease?is increasing with the world's population aging.It was recorded in Compendium of Materia Medica that Celosiae semen can benefit brain,so this paper studied the inhibitory activity of the compounds isolated from the fried Celosiae semen in vitro.The results indicate that compound 7 had obvious inhibitory activity on AChE,the IC50 was 5.16mM.Compound 6,9,10 had weak inhibitory activity on AchE.The results provide the appropriate test and theoretical basis for the prevention and treatment of Alzheimer's disease.
