This paper consists of two parts:chemical constituents of Lagopsis supina and quality control of Semen Aesculi.1 Study on chemical constituents of Lagopsis supinaLagopsis supina(Steph.)Ik.-Gal.(family Labiatae),is widely distributed throughout in China,The herbs are used as gynecological medicine for the treatment of irregular menstruation,painful menstruation,hemiplegia,among others.Previous phytochemical investigations revealed that only nineteen labdane diterpenoids,two flavonoid glycosides,four phenylethanoid glycosides and seven other compounds have been isolated from this plant.In order to clarify the efficacy material basis,action mechanism and chemotaxonomic significance of L.supina,and provide a scientific basis for its further development and utilization,a systematic study was conducted on the chemical constituents and biological activities of L.supina.The results were listed as below.(1)The research progress on chemical constituents and bioactivities of L.supina has been reviewed.Previous phytochemical investigation indicated that labdane diterpenoids,flavonoids,and phenylethanoid glycosides were the main constiuents of this plant.A wide array of bioactivities including anti-inflammatory,antioxidant,myocardial preservation,improving blood and lymph microcirculation disorders of the crude extract of L.supina have been reported.(2)The chemical constituents from the whole plants of L.supina were separated and putified via various column chromatographic techniques including silica gel,ODS,Sephadex LH-20,and semipreparative HPLC.Forty-three compounds were separated and identified from this herb including ten phenylethanoid glycosides:lagopside A(1),lagopside B(2),leucosceptoside A(3),stachysoside C(4),acteoside(5),stachysoside A(6),stachysoside B(7),isolavandulifolioside(8),cistanoside D(9),jionoside C(10);one phenylmethanoid glycoside:salsaside B(11);twelve flavonoids:5-hydroxy-7,4'-dimethoxyflavone(12),genkwanin(13),chrysoeriol-7-O-?-D-glucoside(14),luteolin-7-O-?-D-glucoside(15),apigenin-7-O-?-D-glucoside(16),kaempferol-3-O-?-D-glucoside(17),isorhamnetin-3-O-?-D-glycopyranoside(18),quercetin-3-O-?-D-glucoside(19),rutin(20),tiliroside(21),kaempferol-3-O-6"-(3-hydroxy-3-methylglutaryl)-?-D-glucoside(22),quercetin-3-O-6"-(3-hydroxy-3-methylglutraryl)-?-D-glucoside(23);four megastigmane glycosides:roseoside ?(24),phlomuroside(25),phlomisionoside(26),bridelionoside B(27);five irioids:ajugoside(28),8-O-acetylharpagide(29),ajugol(30),harpagide(31),antirrinoside(32);three monoterpenoid glycosides:6,7-dihydroxy-dihydrolinalool-3-O-?-D-glucopyranoside(33),betulalbuside A(34),(3S,6E)-8-hydroxylinalool-3-O-?-D-glucopyranoside(35);two phenolic glycosides:2-hydroxy-5-(2-hydroxyethyl)phenyl-1-?-D-glucopyranoside(36),methyl 2-O-?-D-glucopyranosylbenzoate(37);two ester glycosides:1-O-coumaroyl-?-D-glucopyranose(38),1-O-caffeoyl-?-D-glucopyranose(39)and four other compounds:adenosine(40),sucrose(41),phytol(42),daucosterol(43).Lagopside A(1)and lagopside B(2)were identified as new compounds.This is the first report of compounds 9,11,22-24,27 and 33 from the Labiatae family,while compounds 3-4,6-8,10,12-21,25-26,28-32,34-42 were isolated from the Lagopsis genus for the first time.(3)The nitric oxide(NO)inhibitory activities of some of the isolated compounds were evaluated by LPS-stimulated BV-2 cells in vitro and seventeen compounds exhibited inhibitions of NO production.(4)The preliminary chemotaxonomic significance of the isolated compounds was summarized on the basis of literature data,especially the distribution of these compounds within the labiatae family.2 Study on quality control of Semen AesculiSemen Aesculi is the dried ripe seeds of Aesculus chinensis Bge.,Aesculus chinensis Bge.var.chekiangensis(Hu et Fang)Fang or Aesculus wilsonii Rehd.(family:Hippocastanaceae).It is a commonly used traditional Chinese medicine documented in Chinese Pharmacopoeia(2015 Edition).Escin is the main component in Semen Aesculi,and its sodium salt has been widely used in clinical practice.But only the content of escin la was used to control the quality of Semen Aesculi in ChP 2015 so far.Therefore,it is necessary to improve the quality evaluation system to ensure the safety and efficacy of Semen Aesculi.This study focus on the establishment of quantative and qualitive method to control the quality of Semen Aesculiand its powder-injection.The results are listed as below.(1)Research progress of chemical constituents,bioactivities,and quality control of Aesculus plants have been reviewed.The main chemical constituents are escin,flavonoids,tannins,and organic acid in genus Aesculus.A wide array of biological activities including antiedematous,anti-inflammatory,antitumor,improving intravenous tension and so on.HPLC is the common methed of quality control for Semen Aesculi,component analysis of Semen Aesculi has not been reported.(2)A UFLC-DAD method for content determination of escin ?a ang ?b was established.The precision,repeatability,stability and accuracy of the method for content determination are good and can be used for the quality control of Semen Aesculi.In addition,the simultaneous determination of escin ?a and escin ?b in 12 batches of Semen Aesculi were carried out by using the developed UFLC-DAD.(3)The MS fragmentation behaviors of escins were summarized by the LCMS-IT-TOF analysis.84 compounds were tentatively identified in Semen Aesculi and 17 compounds were tentatively identified in powder-injection of Semen Aesculi,respectively.
