The experimental study of sargassum fusiforme seleno-polysaccharide on human hepatocellular carcinoma HepG2 cell growth inhibition and apoptosis and discusses its mechanism and the way.In the experiment,MTT showed sargassum fusiforme seleno-polysaccharide on HepG2 cell has certain cytotoxicity,and with the sargassum fusiforme seleno-polysaccharide concentration increases inhibition enhances,the HepG2 cell IC50 32.47ug/ml.Through the inverted microscope observation in direct sargassum fusiforme seleno-polysaccharide human hepatocellular carcinoma HepG2 cells after 48h,negative control group cells arranged relatively close together,cell morphogenesis is complete and growing well.With the concentration increased cell growth density gradually thinning,cell shrinkage,high concentration group most of cell disruption,cell morphogenesis is complete,adherent to reduce.Description of sargassum fusiforme seleno-polysaccharide on HepG2 cell is apoptosis,and it is dose-dependent.Observed under fluorescence microscopy after Hoechst33258 staining,normal cells showed uniform blue fluorescent nuclei,and apoptotic cells nucleus showed a dense dense or chunky concentrated dye fluorescence.The experimental results show that,sargassum fusiforme seleno-polysaccharide role for HepG2 cells after 48h,the negative control group the results showed that HepG2 cells were uniform chromatin,nuclear morphology rules,in dosage group nuclei fragmentation,high dosage group of nuclei showed a dense,this suggests that sargassum fusiforme seleno-polysaccharide can result in HepG2 cell morphological change,and the cells change with the increase of dosage,appeared phenomenon of apoptosis.The experimental use of propidium iodide(PI)single staining by flow cytometry analysis showed that different concentrations of sargassum fusiforme seleno-polysaccharide role of HepG2 cells after 48h,have obvious apoptosis peak,each cell apoptosis rate were(2.44±0.019)%,(39.18±1.90)%,(42.77±3.02)%,as the drug concentration increased,further description of sargassum fusiforme seleno-polysaccharide inhibition effect on HepG2 cells and inducing tumor cell apoptosis related.Based on the experiment on above,this experiment also examines the sargassum fusiforme seleno-polysaccharide induces apoptosis in human hepatocellular carcinoma HepG2 cell apoptosis.By confocal laser scanning microscopy for detection of mitochondrial membrane potential Ca2+,RUS,change,different concentrations of sargassum fusiforme seleno-polysaccharide on HepG2 48h,mitochondrial membrane potential of fluorescence intensity were 27.66±4.50,15.69±3.30,and 10.89±4.49.The Ca2+ of fluorescence intensity were 7.03±0.81,9.96±1.34,and 18.63±5.13.The ROS of fluorescence intensity were 4.65±0.29,5.56±0.27,and 7.14±0.33.The result show that sargassum fusiforme seleno-polysaccharide can reduce uman hepatocellular carcinoma HepG2 cells mitochondrial membrane potential,and are dose-dependent.With the increase of the sargassum fusiforme seleno-polysaccharide doses of Ca2+ concentration increased.And the ROS contention is increasing with the sargassum fusiforme seleno-polysaccharide concentration increasing,and are dose-dependent.To sum up,this thesis through in vitro experiments demonstrate the sargassum fusiforme seleno-polysaccharide can induce apoptosis in human hepatocellular carcinoma HepG2 cell apoptosis,and mitochondrial pathways are sargassum fusiforme seleno-polysaccharide induces apoptosis in human hepatocellular carcinoma HepG2 cell apoptosis may be one way. |
