| This experiment around the sargassum fusiforme seleno-polysaccharide inhibit the growth of human gastric cancer SGC-7901 cells study,aims to explore the action on the sargassum fusiforme seleno-polysaccharide can inhibit the growth of gastric cancer cells.Determined by MTT colorimetry to detect the sargassum fusiforme seleno-polysaccharide effect on SGC-7901 cell activity of the experimental results showed that sargassum fusiforme seleno-polysaccharide has cytotoxic effect,can inhibit the growth of SGC-7901 cells,and with the increase of the unit dosing amount of thick,the higher the relative mortality of SGC-7901 cells,IC50 alue concentration of 34.97 μg/ml.After sargassum fusiforme seleno-polysaccharide affecting on human gastric cancer SGC-7901 cells in 48 hours,use inverted microscope to observe each cell morphological ehanges.With sargassum fusiforme seleno-polysaceharide unit dosage inereasing,the density of growth cell is reduced,the 1 number of adherent cells is in decrease,cells became round and broke.Similarly,Hoechst33258 stain the SGC-7901 cells that was affeetde by sargassum fusiforme seleno-polysaccharide in 48 hours.Use a fluorescence microscope to observe the changes in the state of cells in each group.The experimental results show that,sargassum fusiforme seleno-polysaccharide role for SGC-7901 cells after 48h cell shape changes with the increase of dosage,appeared phenomenon of apoptosis,the negative control group the results showed that the SGC-7901 cell chromatin uniform,nuclear morphology rules,in dosage group nuclei fragmentation,high dosage group of nuclei showed a dense dense,this suggests that sargassum fusiforme seleno-polysaccharide can result in SGC-7901 cell morphological change.The experimental use of propidium iodide single staining by flow cytometry analysis showed that different concentrations of sargassum fusiforme seleno-polysaccharide role of SGC-7901 cells after 48 h,have obvious apoptosis peak,each cell apoptosis rate were low dose group(7.38±0.32)%,middle dose group(11.45±0.45)%,high dose group(23.81±1.16)%.As the drug concentration increased,further description of sargassum fusiforme seleno-polysaccharide inhibition effect on SGC-7901 cells and inducing tumor cell apoptosis related.On the basis of the above experiments,this study also explored the sargassum fusiforme seleno-polysaccharide induced the essence of human gastric cancer cells apoptosis.This experiment detected the change of mitoclhondrial membrane potential by laser confocal microscopy.Different concentration of sargassum fusiforme seleno-polysaccharide effect on SGC-7901 after 48h,mitochondrial membrane potential fluorescence intensity is respectively:low dose group(36.95±0.41),middle dose group(27.47±0.31)and high dose group(18.06±0.20).This experiment detected the change of intracellular reactive oxygen species by laser confocal microscopy.Different concentration of sargassum fusiforme seleno-polysaccharide effect on SGC-7901 after 48h,intracellular reactive oxygen species fluorescence intensity is respectively:low dose group(5.90±0.09),middle dose group(8.42±0.27)and high dose group(12.64±0.76).This experiment detected the change of calcium ion concentration by laser confocal microscopy.Different concentration of sargassum fusiforme seleno-polysaccharide effect on SGC-7901 after 48h,calcium ion concentration fluorescence intensity is respectively:low dose group(9.10±0.06),middle dose group(11.27±0.07)and high dose group(23.59±0.42).After results was analysised,each group have significant statistical significance(P<0.01).The results show that the sargassum fusiforme seleno-polysaccharide induction of human gastric cancer SGC-7901 cells apoptosis through mitochondrial pathway:selenium polysaccharide function to the cell,the sargassum fusiforme seleno-polysaccharide as the dosage increased mitochondrial membrane integrity are destroyed,decrease the mitochondrial membrane potential and active oxygen content increases,calcium ion concentration increased sharply,so that the mitochondrial membrane channel open hole,resulting in cell apoptosis.To sum up,this thesis through the in vitro experiments proves that the sargassum fusiforme seleno-polysaccharide can induce gastric cancer SGC-7901 cell apoptosis,and mitochondrial pathway is the sargassum fusiforme seleno-polysaccharide induction of human cancer SGC-7901 cells apoptosis may be one way. |
