| Crinum asiaticum L.belong to herb of Amaryllidaceous Crinum Genus,crinum is one of the most important medicinal plants,it has a great medicinal value.In this test we use the seed of crinum to study its falconoid;We do the research of extraction.We use the technique of ultrasonic assisted extraction to optimize the Solid-liquid ratio?the concentration of ethanol?extraction time and temperature through the response surface to make sure the best extraction.We get the results that:Solid-liquid ratio is 1:28,extraction time is 47min,extraction temperature is 64?.We measure the extraction rate is 1.61%,it's closed to the predictive value of 1.65%.We study the effect of four kinds of macro porous resin to the purification of Crinum falconoid through static adsorption and desorption.And AB-8 is the best,through the AB-8 static adsorption and desorption we get the best dynamic elution condition is that sample concentration of 2mg/mL,the sample rate of lmL/min,the sample volume 6BV,95%of ethanol concentration,the elution rate of lmL/min,elution amount of 5BV.We collect the eluent,concentrate freeze-dried and get the crude of Crinum falconoid.Make the crude of Crinum falconoid through polyamide column to get the further purification;at last we get the boutique of crinum asiaticum L.falconoid.Application LC-Q/TOF-MS/MS analysis of the chemical composition of flavonoids in Crinum asiaticum L.,identified the major peaks of flavonoids boutique MS total ion current combining literature,20 peaks were identified,seven major components,the peak 1 is Asiaticumine A,peak 3 is Asiaticumine B,peak 4 is Latifoliumin C,peak 5 is Ungeremine,peak 18 is 2,5 '-O-nitrosouridine,peak 19 is Lycorenan-7-one,peak 20 is Crinamine,but no one was flavonoids,remaining was unknown.But hydrochloride-the color reaction of magnesium powder,ferric chloride,sodium hydroxide and other flavonoids are positive,indicating Crinum boutique contains flavonoids,but has not yet been identified. |
