Polysaccharides have anti-tumor,lowering blood pressure,improve the immune system and many other effects,as a substance that extracs from a natural plant which basically no side effects,but there is a vast pharmacological research value and application prospect.In this paper,Crinum polysaccharide as the research object,a clear optimum extraction of Crinum polysaccharides,grasp the extraction process of Crinum polysaccharides,testing physical and chemical properties of Crinum delicate polysaccharides and sophisticated understanding of Crinum polysaccharide on tumor cells in vitro.The main results are as follows:The experiment using ultrasonic assisted extraction,and single factor solid-liquid ratio,ultrasonic time and ultrasonic temperature three factors and four levels of study,then the experimental design using Box-Behnken,then through the use of software to get optimal results extraction conditions.Finally get optimum extraction conditions for liquid-solid ratio was 11:1,ultrasonic time of 58min,a temperature of 65?.The experiment Crinum pending extract water line extraction and ethanol precipitation to obtain Crinum crude polysaccharide,then in addition to protein,in addition to color,to detect alcohol precipitation step to get the maximum amount of polysaccharide group was divided into 30%,and then get through the dialysis Monju Lan semi refined polysaccharide.Last DEAE-52 component is detected 0.2mol/LNaCl of peaks can be better,dialyzed and lyophilized again to give purified Crinum polysaccharide.Glucan polysaccharide had been refined after G-200 and HPLC purity detection of Crinum refined polysaccharides,the experimental result prove that the method has the ability to extract the Crinum refined polysaccharide and the content of Sugar is 61.2%.This experiment measured the solubility of the polysaccharide,the polysaccharide solution can be assisted by ultrasonic salt solution,Fehling reaction was negative,Molish reaction with a purple ring,ninhydrin reaction was negative,ferric chloride was negative,Biuret reaction was negative,Phenol sulfuric acid reaction was Positive.The content of sugar aldehyde was 29.8%and the content of sulfuric acid base was 5.06%sugar content of sulfuric acid aldehyde carbazole method.The above experiments show Crinum polysaccharides in liquid-solid ratio was 11:1,ultrasonic time of 5 8min,temperature of 65?,under the conditions have the maximum extraction rate.After a series of separation and purification crinum polysaccharides can be more pure Crinum refined polysaccharides.Crinum refined polysaccharide containing sulfate groups and aldehyde.
