Research On The Chemistry To Neutral-Strong Alkaloid From Crinum

Crinum has come from Amaryllidaceae white Crinum which is a perennial herb,Usually prefer to live in coastal and river sand and other regions.In addition,it has been cultivated in many places,such as in southern regions of China,tropical and subtropical regions.Crinum is rich in alkaloids.Initially,it often is used as a drug with Crinum leaves and rhizomes in the crowd,It have visible efficacy that clear fire detoxification and eliminate bruising efficacy.Extracts obtained from Crinum bulbs can be well suppressed cell activity which human lung cancer cells,colon cancer cells and leukemia etc.But the reports are very few.Crinum leaf extract has anti-tumor activity,inhibition of human lung cancer cells(A549),colon cancer cells(LOVO),T-cell leukemia cell growth activity in previous discoveries.The chloroform extract is having a significant anti-tumor effect,can inhibit the proliferation of tumor cells NCI-H460,and cause apoptosis,cell cycle arrest in G1/S phase.Study on Chemical Constituents activity with the seeds of Crinum alcohol extract in this experiment,Research on chemical activity of ethanol extract of the Crinum seeds provide important theoretical guidance and experimental basis for further development and clinical application of Crinum through extraction and purification.To optimize the ethanol extraction process of total alkaloids from Crinum,the single factor were studied first,such as ethanol concentration,extraction temperature,and time etc.Then,we designed orthogonal experiment which is based on the results of single factor,it is concluded that the best extraction method of Crinum alkaloids is 85%ethanol concentration,65? extraction temperature,extracted in 3h.According to the experimental conditions which are extracted Crinum alkaloids from the orthogonal experiment,we deal with the total alkaloid.We deal with the ethanol extract of total alkaloid which are obtained from Crinum,and adjust pH=7?8 to separate the strongly basic alkaloids.After the moderate base extractum was separated by silica gel column chromatography,collected the detection of each fraction,each fraction collection and detection,when the reaction of alkaloid fractions was positive,the SPE column were used of separation,purification and mass spectrometry detection.LC-MS of detailed analysis,screening of the mobile phase and absorb the liquid conditions such as wavelength,and the methodology,the results show that the effect of the precision,stability and repeatability is good,and then mass spectrometric detected each fractions,we are speculated the structure of sample which combined with reference books and spectral library,and to inferred five structures of the compounds which it can be existed,it respectively that Uridine,Hippeastrine,Lycorenan,Crinasiatine,Tazettine.
