This paper preliminarily studies the primary and advanced Structural Analysis of the purified Polysaccharides from Crinum asiaticum.The anti-tumor activity of polysaccharides also been examined.The paper aims at revealing the structure information of Crinum polysaccharide and whether it has anti-tumor activity or not,in order to find a new basis to the research of the anti-cancer effect of Crinum polysaccharide.The purified Crinum polysaccharide not contains proteins and nucleic acids,and the relative molecular weight is 7.3×105Da.UV scanning spectrum shows that the refined Crinum polysaccharide solution without absorption at 260 nm and 280 nm,which explains that refined Crinum polysaccharides have no proteins and nucleic acids.Through gas chromatographic analysis,the monosaccharide composition of Crinum polysaccharide is mainly composed of Rha,Glc,Gal,Sor(the molar ratio,1:4.74:1.66:61.61);Infrared spectrum analysis results show that the orchid refined Crinum polysaccharides contains typical characteristic absorption peak,the sugar ring form is pyranose ring containing uronic acid and with rhamnose characteristic absorption peak,?-glycosidic linkage;The backbone of polysaccharides chain is connected mainly by(1?2),(1?6),(2?3)bond,and did not have(1?4)bond.Congo red experiment shows that Crinum polysaccharide has a spiral structure with Congo red complex,and has the characteristic reactions occur Congo red triple helix;I2-KI reaction result indicates that Crinum polysaccharide may has many side chains and long branches;A method has been developed to give topography of Crinum polysaccharide by atomic force microscopy(AFM).The result manifests that Crinum polysaccharide has a multi-branched structure and a variety of different linkages between adjacent monosaccharides,which compose the small rings and the helical structure;From the images,the single chain of Crinum is 200~300nm in diameter and 600~900nm in length,which illustrates that the sugar chain is not a single but formed by winding a plurality of shares;Meanwhile,it can be clearly seen from the high-resolution image that there are many polysaccharide particles in the gaps between the mesh and the rod-like,and the diameter is about 42.56nm;The result of powder X-ray diffraction shows that the polysaccharide contains few crystalline polysaccharide or polysaccharide complexes and has poor crystallinity;The result of particle size distribution indicates that the average particle size of polysaccharides Crinum is 470.12?m.We also have studied the primary anti-tumor activities of Crinum polysaccharide by means of MTT,the results prove that Crinum polysaccharide has activities to the cancer cell(HepG2),with a concentration dependant ration.IC50 of 72h is 128.07?g/ml.Structural analysis shows that purified Crinum polysaccharide has a triple helix,the sugar chain containing(1?3),(1?6)glycosidic linkages,?-glycosidic linkage,pyranose ring of sugar,urolic acid groups and the molecular weight is within 105~106Da,which may contribute to the anti-tumor activity of Crinum polysaccharide.In summary,Crinum purified polysaccharide has anti-tumor activity,is another active substance in genus Crinum plant except alkaloid,which finds a new basis for the anti-cancer effects of Crinum.
