Study On The Chemical Constituents Of Sesquiterpene From Solid-stated Fermented Antrodia Camphorata

Antrodia camphorata is a kind of rare and unique Food and Medicine fungus in Taiwan.Its main active ingredients contains ubiquinone compounds,terpenes,maleic acid and succinic acid derivatives and benzene derivatives.The sesquiterpene compounds antrocin which isolated from the fruiting body of wild Antrodia camphorata has a good anti-cancer activity,but whether the sesquiterpene compounds existed in solid-stated fermented Antrodia camphorata has not been reported.So in this study,the sesquiterpene compounds were isolated.from solid-stated fermented Antrodia camphorata.In this study,the solid-stated fermented Antrodia camphorata was extracted with 95% ethanol,and then the extract was suspended in H2 O and partitioned with ethyl acetate.The ethyl acetate soluble fraction was concentrated under reduced pressure to obtain an ethyl acetate extract.And then the ethyl acetate extract was separated by column chromatography on silica gel,Sephadex LH-20,ODS,and semi-preparative HPLC column chromatography and other methods.Finally,the structure of the compounds were identified by IR,MS,1H NMR,13 C NMR and 2D-NMR spectroscopic.The final study as follows:Three sesquiterpene compound: cadinol(1),nerolidol(2)and a new sesquiterpene 1,7,7-trimethyl-3-hydroxymethyl-bicyclo [4,4,0]-2-decene-2-carbaldehyde(5).Two maleic acid and succinic acid derivatives were also obtained: Antcin A(3)and Antrodin B(4),and they have been reported in the research about Antrodia camphorate.Compound 5 is a new compound.The compound 1 and 2 are separat-ed from Antrodia camphorate for the first time.Antrodin A which can effectively suppress HCV,antcin B can selectivly supress LLC tumor cell lines,cadinol can be used in dendritic cells-based immunotherapy for cancer,nerolidol has antinociceptive and anti-inflammatory effects.So presumably the solid-stated fermented Antrodia camphorata having anti-hepatitis C virus,anti-cancer,antinociceptive and anti-inflammatory activity,the solid-stated fermented Antrodia camphorata has a great development prospects.The syudy provides a more theoretical basis for the medical value of solid-stated fermented Antrodia camphorata,as well as the further development of Antrodia camphorata.
