| ObjectivePart one.This article reveals the influence of different origin on the effective composition of Zhejiang fritillary by studying the effects on the main effective components of original areas and Qingyuan region of Zhejiang Fritillaria decoction pieces.Part two.Through the construction of chemical composition fingerprint chromatogram of total alkaloids of the 15 batch of Qingyuan Fritillaria thunbergii and the 18 batch of Ningbo Fritillaria thunbergii,it determines the fingerprint common pattern and analyses the similarity,thus providing a reference for the quality control of Zhejiang fritillaria.Part three.Through the research on identification and content determination of Zhejiang fritillaria fresh cut slices,it establishes quality standards of Zhejiang fritillary fresh cut slices and provides experimental basis for researches on its quality control.MethodsPart one.The effect of Qingyuan origin on Zhejiang fritillary1.The HPLC-ELSD method was used to study the effect of Qingyuan origin on Zhejiang Fritillaria alkaloids with peimine,peiminine,peimisine as control sample.2.Using the HPLC-ELSD method,the contents of peimine,peiminine,peimisine of Fritillaria thunbergii pieces from different origins were measured.Part two.Research on the chemical composition fingerprint chromatogram of total alkaloids of Zhejiang fritillary from Qingyuan and Ningbo origin.With peimine,peiminine,peimisine as control sample,the HPLC-ELSD method is used to establish and study the chemical composition fingerprint chromatogram of total alkaloids of Zhejiang fritillary from Qingyuan,and all the chromatograms of 20 batches of samples are obtained by using the method of similarity analysis.Part three.Research on the quality standard of Qingyuan fritillary fresh cut slices.1.Content Determination Using the HPLC-ELSD method,the content of peimine,peiminine and peimisine are determined.2.Identification condition on TLC.The thin layer plates of silica gel are used with peimine and peiminine as control sample.3.Inspection.Determining items of sample that include moisture,Heavy metal,ashes,Pesticide residues and water extracts.ResultsPart one.Through the comparison of the HPLC-ELSD fingerprint chromatograms of Zhejiang fritillary fresh cut slices from different origins,the effective composition contents and the extract contents,the experimental results show that the difference of the quality is very sharp,the migrations of Zhejiang fritillary not only affect the main effective components,but also the contents of peimine,peiminine and peimisine.Part two.The chemical composition fingerprint chromatogram of total alkaloids of the 15 batch of Qingyuan Fritillaria thunbergii and the 18 batch of Ningbo Fritillaria thunbergii is established and the fingerprint common model is determined.Similarity analysis method is stable and reliable,has good repeatability.Part three.1.Zhejiang fritillary bulb powder has distinct microscopic characteristics,such as anticlinal walls of the epidermal cells of renju shaped thickening,visible pores,and there are 4~5 accessory cells;Zhejiang fritillary bulb extract of the same position shows silica gel plate number,size,color spots.All of these show that all the samples have good similarity.2.The content of the moisture,ashes,Sulfur dioxide and water extracts meet the standards of Fritillaria thunbergii.[Ch.P(2010)Vol 1,274].3.In the assaying,the precision,reproducibility and recovery all meet the requirements.The content range of peimine,peiminine and peimisine from 13 batches of samples are0.061%~0.082%、0.042%~0.053%、0.031%~0.043%.4.13 batches of zhejiang fritillary fresh cut slices,666,five nitrobenzene chloride,the content of DDT in 0.014 mg·kg-1~0.034 mg · kg-1,0.001 mg·kg-1~0.002 mg·kg-1,0.028 mg·kg-1~0.062 mg·kg-1 range,conform to "green trade standards of importing medicinal plant and meet its preparation of related standards(666,five nitrobenzene chloride,DDT residues are for 0.1 mg·kg-1)or less.5.Reserch on the 13 batches of zhejiang fritillary fresh cut slices samples shows heavy metals and trace elements content in sample elements As,Cd,Cu and Pb content range of 0.01~0.22μg·g-1,0.19~0.29μg·g-1,1.43μg·g-1~3.47μg·g-1,0.01~0.35μg·g-1 Hg undetectable,consistent with "green trade standards of importing medicinal plants and their preparation for the requirements of the heavy metals and trace elements of traditional Chinese medicine.ConclusionsPart one.The migration of Zhejiang fritillary to Qingyuan has great affect on both the main effective components and its content.Part two.This experiment established the chemical composition fingerprint chromatogram of total alkaloids of the 15 batch of Qingyuan Fritillaria thunbergii and the 18 batch of Ningbo Fritillaria thunbergii and determined the fingerprint common model.Part three.It also established the quality standards of Fritillaria thunbergii.The identification and content determination method has strong specificity and it is simple,accurate and reliable operation,good repeatability,which can effectively control the quality of the Zhejiang fritillary slices. |
