| Objective:To observe the effect of moxibustion on activating enzyme NIK,transforming growth factor beta 1 Experimental RA rabbit synovial tissues(TGF-β1),nuclear factor kappa Bp65(NF-κBp65)and vascular endothelial growth factor(VEGF)on the expression of: To observe the effect of moxibustion on NF-κB/TGF-β1 signaling pathway and VEGF to investigate the mechanism of Moxibustion in the treatment of RA.Methods:30 Japanese white rabbits(referred to as "rabbit"),only half of the male and female,weight of 2.5 + 0.25 kg.One week before domestication.Randomly divided into blank control group(referred to as "blank"),RA model group(referred to as the control group),moxibustion treatment group(referred to as treatment group),each group of10.The control group and the treatment group were treated with Freund’s complete adjuvant(FCA)method,and the dosage was 0.5ml/Kg.Blank group without any treatment.At seventh days,with Moxa Moxibustion treatment group rabbit bilateral "Shenshu " and " Zusanli ",two points each 5 strong,once a day,alternately on both sides.6 days for a course of treatment,a total of 18 days,a day off between the two courses.Blank group and control group were not treated with moxibustion.After twenty-first days of treatment,the animals were sacrificed and the synovial tissue and synovial fluid were taken.Enzyme linked immunosorbent assay(Elisa)was used to detect the contents of IL-6,TNF-α and VEGF in synovial fluid of rabbits in each group.The expression of NF-κBp65,NIK,TGF-β1 protein in synovial tissues was determined by immunohistochemical staining SP methodResult:1 Compared with the blank group,the content of NF-κBp65 protein in the control group was significantly higher than that in the control group(P<0.01).After treatment,compared with the control group,the expression of NF-κBp65 protein decreased in the treatment group(RA),which was statistically significant(P<0.05).2 Compared with the blank group,the content of TGF-β1 protein in synovial tissue of the control group was significantly higher than that of the control group()(P<0.01).After treatment,compared with the control group,the expression of TGF-β1 protein in the synovial membrane of RA rabbits in the treatment group was significantly lower than that in the control group(P<0.01).3 Compared with the blank group,there was no significant difference in the expression of NIK protein between the control group and the treatment group(P>0.05)in the synovial tissue of rabbits(RA)4 Compared with the control group: the control group and the treatment group RA content of rabbit synovial fluid IL-6,TNF-α,VEGF increased significantly after treatment(P<0.01);content of these cytokines in synovial fluid was significantly decreased than model group(P<0.05).Conclusion:1 Moxibustion "Shenshu" and "Zusanli" can reduce the RA of rabbit synovial tissue NF-κBp65,TGF-β1protein content,significantly reduce the expression of rabbit synovial tissue strongly positive influence of NF-κB/TGF-β1 signaling pathway,and inhibiting vascular pannus of newborn.2 Moxibustion "Shenshu" and "Zusanli" can inhibit the experimental RA rabbit synovial inflammation tissue IL-6,TNF-α,VEGF expression and effect of NF-κB/TGF-β1 signaling pathway in the development and expression of VEGF,reduce the formation of pannus,RA joint inflammation,with clear anti-inflammatory and immune effect,may is the internal mechanism of moxibustion therapy for RA. |
