| Objective:Fresh bamboo leachate(Xianzhuli)is Gan and Cold,has the function of removing pathogenic heat and resolving phlegm.It can increase the bronchial ciliary movement,so that the sputum can be discharged from the trachea and bronchia.It is especially effective to the thick and sticky sputum which is difficult to spit out.The main dosage forms clinically used are Xianzhuli Oral Liqiud,Zhuli granule and Zhuli capsules,etc.,they have the effects of clearing heat and resolving phlegm,but there are few reports on the comparative study of the efficacy of different dosage forms.In this study,SD rats were used.The rat model of chronic bronchitis was reconstructed by modified smoking method.The effects and mechanisms of Zhuli granule on the protection and improvement of lung injury in chronic bronchitis model rats were studied,as well as the comparative study of Zhuli Granule and Xianzhuli Oral Liquid was performed too.We hope this research will richen the clinical applications of different fresh bamboo leachate dosage forms.Methods : SD rats were randomly divided into Normal group,Model group,Granule group and Oral Liquid group.The rat model of chronic bronchitis was established by modified smoking method(cigarettes smoking).In the course of smoky modeling,through internal experiments,Zhuli Granule and.Xianzhuli Oral Liqiud were used by gavage to intervene the progress of chronic bronchitis.The research methods included pathology,transmission electron microscope,spectroscopic analysis and enzyme-linked immunosorbent assay.To observe the lung tissue morphology,lung tissue and vascular damage,lung antioxidant capacity,secretion of pulmonary inflammatory mediators and so on,and the preliminary analysis and comparison of the main active ingredients of Zhuli Granule and Xianzhuli Oral Liquid.Finally,to investigate the curative effect and mechanism of Zhuli Granule and Xianzhuli Oral Liquid on chronic bronchitis model rats.Results:(1)Changes of lung histopathology: Zhuli Granule and Xianzhuli Oral Liquid could alleviate the pathological changes of lung tissue in model rats with chronicinflammation caused by smoke and reduce the structural damage of lung tissue.Under the observation of light microscope and electron microscope,there was no obvious pathological damage in Normal group,and the pathological damage of the Model group was significant.The results showed that both Granule group and Oral Liquid group could obviously reduce the pathological damage and the effect of Granule group was better than Oral Liquid group.(2)The condition of lung injury: The content of serum NO in model rats was significantly decreased and the HP content in lung tissue was significantly increased.Zhuli granule and Xianzhuli Oral Liqiud could significantly promote the secretion of NO,so that the content of NO in serum was significantly higher than that in Model group.At the same time,Zhuli granule and Xianzhuli Oral Liqiud could significantly reduce the lung tissue homogenate HP content,so the content of lung tissue was significantly lower than that in Model group.There was no significant difference between Granule group and Oral Liquid group(P> 0.05).(3)The antioxidant capacity of lung tissue: The antioxidant capacity of lung tissue in rats with chronic inflammation caused by smoke was weakened.The activity of antioxidant enzymes SOD in the lung tissue homogenate was significantly decreased and the content of MDA(the lipid peroxidation product)was significantly increased.Zhuli Granule and Xianzhuli Oral Liqiud could improve the antioxidant capacity of lung tissue in model rats with chronic bronchitis,showed a clear intervention effect.Compared with the Modle group,the SOD activity of the lung tissue homogenate was significantly increased and the content of MDA was significantly decreased in the Granule group and Oral Liquid group.The effect of Granule group was more obvious than Oral Liquid group,but there was no significant difference between the two groups(P> 0.05).(4)The secretion of pulmonary inflammatory mediators: The levels of inflammatory mediators TNF-? and IL-6 in bronchoalveolar lavage fluid(BALF)in model rats with chronic inflammation were significantly increased.Both Zhuli Granule and Xianzhuli Oral Liquid could reduce the secretion of inflammatory mediators in bronchoalveolar lavage fluid.The content of TNF-? and IL-6 in theGranule and Oral Liquid groups were significantly lower than that in Model group,and the effect of Granule was more obvious.However,there was no significant difference between these two groups(P 0.05).(5)Analysis of active ingredients: Total phenol is one of the important active ingredients in fresh bamboo leachate.The results showed that the total phenol content of Zhuli Granule was much higher than that of Xianzhuli Oral Liqiud.Conclusion:.(1)Zhuli Granule and Xianzhuli Oral Liqiud have certain protective effect on the lung tissue and airway injury caused by smoke inflammation in chronic bronchitis model rats.The possible mechanisms included:(1)Zhuli Granule and Xianzhuli Oral Liquid have some protective effect on the lung tissue and airway injury in chronic bronchitis model rats caused by smoke.(2)Zhuli Granule and Xianzhuli Oral Liqiud can enhance the ability of anti-oxidative injury in chronic bronchitis model rats caused by smoke.(3)Zhuzhi Granule and Xianzhuli Oral Liqiud can reduce the secretion of inflammatory mediators in the lung tissue of chronic bronchitis model rats caused caused by smoke and reduce the inflammatory injury.(2)In many aspects,the effects of Zhuli Granule is better than that of Xianzhuli Oral Liquid,which may be related to the different processing and lead to the differences in their main active ingredients.Both the UV method and the phenolic phenol method showed that the content of total phenol in the Zhuli Granule was significantly higher than that of Xianzhuli Oral Liquid.But between them,there is no statistically significant difference,which indicate that fresh bamboo leachate have other important active ingredients involving in the function of removing pathogenic heat and resolving phlegm. |
