Research Onpharmacodynamics And Mechanism On Chronic Bronchitis Of Danxian Kechuangning Capsules

Purpose:Chronic Bronchitis(CB),is a public recognized health threaten disease glo-bally,the main clinical characteristics is recurrent cough,sputum,or common respiratory system diseases with wheezing.With the deepening of the disease,the frequency and severity of chronic bronchitis will also increase.Eventually it will lead to chronic obstructive pulmonary emphysema or corpulmonale,which will seriously affect the patient’s life quality and even life span.Danxian Kechuanning Capsules are composed by five drugs:pulvis fellis suis,pinellia,dried tangerine or orange peel,poria cocos and epimedium.With the function of This article mainly expounds the research of Danxian Kechuanning Capsules’ pharmacodynamics and mechanism on chronic bronchitis.Method:1、Use experiments of rat swelling feet caused by egg white to study the anti-inflammatory action of Danxian Kechuanning Capsules.2、With the method of mice cough caused by ammonia,phenol red excretion in mice,guinea pigs breath method,determination of mice cough incubation period of cough and five minutes,gas tube filling lotion,content of phenol red in the incubation period of asthma guinea pigs to explore thismedicine’s curative effect for cough,phlegm,wheezing and other respiratorydisease3、Use improved smoked method to simulate the model of chronic bro-nchitis.Puts the rats into a customized 3 feet smoked chamber.Smoke the chamber with 50g wood shavings,50g sawdust,and 50g tobacco,and do it twice a day,45 minutes each time,and last 45 days.Raise the controlgroup in a normal smoke-free environment.4、The 54 SD rats,male and female combination,randomly dividedinto6 groups,9 in each group.Remember the 6 groups as:Blank Control,ModelControl,High Dosage,Middle Dosage,Low Dosage,positive Control group(DXM).Dosage delivery system using oral lavage.Begin to drug them from2 days after building mold.Give the same amount of normal saline to blankcontrol group and model control group,once per day,for 45 days.5、Observe and record each group rats before and after building model and general condition before and after the administration,including mental state,activity,breathing,fur color and lustre and the change of body weight6、After execution of the rat,open the chest and isolate full lung tissue,and visual inspect the two lungs tissue morphology.Then use the optical microscope to observe the lung tissue morphology7、By enzyme-linked immunosorbent assay(ELISA)in serum and lung tissue of rats with chronic bronchitis and inflammatory mediators IL-8、MMP-9、,and the content of ICAM-1Result:1、The research of Danxian Kechuanning Capsules’ anti-inflammatory shows that this medication can suppress rat swelling feet,comparing to the Blank Control Group2、The research of Danxian Kechuanning Capsules’ relieving cough shows that this medication can expand the incubation period of cough and de-crease the cough times of mice,comparing to the Blank Control Group.3、The research of Danxian Kechuanning Capsules’ eliminating phlegm shows that this medication can increase the excretion of mice’s tracheal secretions,comparing to the Blank Control Group.4、The research of Danxian Kechuanning Capsules’ relieving cough shows that this medication can expand the incubation period of cough and decrease the tracheal contraction obviously of cavies and suppress the cavies’ tr-acheal spasm caused by Histamine diphosphate,comparing to the Blank Co-ntrol Group5、Improved smoked method to simulate the chronic bronchitis model results show that the model control group rats with extension of smoking time,there are different level of coughing and deepen breathing,deepen color of nose and mouth mucosa secretions,being tired and laying still,mental tired,eating less,and with dried yellowed fur.There were no significant differences between groups of rats weight before experiment.After experiment building each smoked group rats’ weight decreased obviously compared with blank control group rats(P<0.01);After the treatment,the mental state of rats from High,Medium dosage groups and positive control group gradually improving,hair glossiness increases,breathing gradually leveled off,nose and mouth mucosa secretion decreased,increased activity,diet increase.Model control group rats’ weight compared with blank control group decreased obviously,the weight of rats from High,Medium dosage and positive control groups increased significantly compared with model control group(P<0.01).6、After treatment,visual inspection the lung tissue morphology shows the result that in the High,medium,low dosage and the positive control groups,some part of the lung tissue are in dark red,and occasionally bleed-ing,which are different from Model control group significantly.thin bronchial mucosa epithelium is incomplete,no obvious expansion and lumen with infl-ammatory cells infiltration.7、Compare to the Model control group,ELISA Method shows that High,Medium,Low,and Positive control group can decrease the content of IL-8、MMP-9、IC AM-1 in rats’ serum and lung(P<0.01 or P<0.05)...
