Objective: Observation and tendons technique of rabbit knee osteoarthritis model in pathological changes of articular cartilage and joint fluid interleukin-1 beta serotonin levels of beta-endorphin cartilage of type ? collagen expression changes,to explore the deep mechanism of tendons technique,which provides new thinking for clinical treatment of knee osteoarthritis.Research methods: The 48 rabbits were randomly divided into blank group,model group,sodium hyalurate group,and "shu jin" technique group(hereinafter referred to as manipulation group),each group of 12.Except the blank group adopt method of articular cavity injection of papain replication model of knee osteoarthritis,give group of rabbit articular cavity injection of sodium hyaluronate sodium hyaluronate treatment,to give technique group rabbit "muscle" technique treatment,once a day.The model group and the normal group were not treated.Each group respectively for 8 weekend,weekend 12 each executed 6 rabbits,macroscopic and the microscopic observation of the knee joint cartilage degeneration,using enzyme-linked immunosorbent method determination of interleukin 1 beta in joint fluid,serotonin,the content of beta-endorphin,using Western blot method-determination of type ? collagen expression in rabbit cartilage.Results:(1)Improve LequesneMG knee level assessment,general morphological and histological observation of articular cartilage score,8 and 12 weekend over the weekend,model group,sodium hyaluronate,technique group compared with normal group,significantly increased,by the single factor analysis of variance,statistically significant(P < 0.05);Compared with the model group,the grade of sodium hyaluronic group was significantly reduced,and there were statistically significant differences(P < 0.05).The comparison of sodium hyaluronic group and technique group was similar,and there was no statistically significant difference(P>0.05).Compared with the 8th weekend in the same group,the score of sodium hyalurate group and technique group was reduced,and there were statistically significant differences(P < 0.05).(2)ELISA method for determination of interleukin 1 beta in joint fluid,serotonin,the content of beta-endorphin,8 and 12 weekend over the weekend,model group,sodium hyaluronate,technique group compared with normal group,the three levels increased significantly,the single factor analysis of variance,statistically significant(P < 0.05);Compared with the model group,the content of sodium hyaluronic acid group was significantly reduced,and there were statistically significant differences(P < 0.05).The content of sodium hyaluronic acid group was comparable to that of the technique group,and there was no statistically significant difference(P>0.05).Compared with the 8th weekend in the same group,the content of sodium hyalurate group and technique group decreased,and there were statistically significant differences(P < 0.05).(3)Western-blot method determination of type ? collagen expression in rabbit cartilage: 8 and 12 weekend over the weekend,model group,sodium hyaluronate,technique group compared with normal group,stripe color becomes shallow,shows that the expression of type ? collagen content decreased significantly;Sodium hyaluronate group,manipulation group and model group were compared,and the color of stripe was significantly increased,indicating a significant increase in the expression of type ? collagen.The comparison of sodium hyaluronate group and technique group showed that the expression of type ? collagen was similar.Compared with the 8th weekend in the same group,the color of sodium hyaluronate group and technique group was darker,indicating an increase in the expression of type ? collagen.Conclusion: It can significantly improve the degeneration of articular cartilage in the osteoarthritis of the rabbit knee,inhibit the secretion of inflammatory cytokines,relieve pain,and enhance the expression of cartilage ? collagen. |
