Effect Of "Shu Jin" Manipulation On Type ? Collagen MRNA Expression In Model Rabbits With KOA

Objective:By observing the "shujin" technique for knee osteoarthritis,knee osteoarthritis,KOA)rabbit cartilage type ? collagen expression and related therapeutic indexes,the effects of exploring "shujin" technique on the mechanism of action of cartilage repair.Methods:There were 50 adult rabbits,2 confirmed models,and the other rabbits were divided into 4 groups of 12.In the treatment group of sodium hyaluronate and the treatment group of sodium hyaluronate by the "shu jin" technique,the contrast between the group and the group was compared between 8 weeks and 12 weeks in the blank group.Observation of the knee joint cartilage degeneration,general form enzyme-linked immunosorbent assay(ELISA)determination of serum inflammatory index,pain index indicators IL-1?,5-HT,?-EP content changes,HE observed dyeing cartilage tissue change,reverse transcription polymerase chain reaction(rt-pcr)method to detect the expression of type ? collagen in cartilage.Results:Improved LequesneMG knee level evaluation,articular cartilage morphological and histological observation: roughly 8,12 weekends,compared with model group,sodium hyaluronate group,the score technique group were reduced(P <0.05);There was no significant change in the score of sodium hyaluronic group(P BBB 0.05).Compared with the 8th weekend in the same group,the score of sodium hyalurate group and technique group decreased(P < 0.05).ELISA method was used to determine the contents of IL-1?,5-HT,?-EP in serum: at the end of week 8 and 12,the contents of sodium hyaluronate group,technique group and model group were significantly reduced(P < 0.05).The content of sodium hyaluronate group was comparable to that of the technique group(P BBB 00.05).Compared with the 8th weekend in the same group,the content of sodium hyaluronate group and technique group decreased(P < 0.05).Cartilage type ? collagen mRNA RealtimePCR method to detect the relative expression quantity: 8,12 weekends,sodium hyaluronate,technique group comparedwith model group,the relative expression of type ? collagen mRNA increased significantly(P < 0.05);Compared with the technique group,the relative expression of type ? collagen mRNA was comparable(P BBB 0 0.05).Compared with the 8th weekend in the same group,the relative expression of the type ? collagen mRNA in sodium hyaluronate group and technique group increased(P < 0.05).Conclusion:The method can improve the expression of cartilage type ?collagen and promote the repair of KOA by improving the degeneration of the cartilage of the KOA rabbit and inhibiting the secretion of inflammatory factors.
