Objective:Measure the change of angiotensin ? and blood viscosity after using ginkgolide B to interfere rats with severe acute pancreatitis.To explore the effect of microcirculation disorder on the incidence of severe acute pancreatitis and the therapeutic effect of ginkgolide B on severe acute pancreatitis.Methods:Seven-two adult Sprague-Dawley(SD)rats,no gender restriction,weighting from 250g±50g,were divided into three groups randomly:negative control group(NC group,n=24),N52021 treating group(BN group,n=24),severe acute pancreatitis group(SAP group,n=24).?Negative control group(NC group):only touch the pancreas three times after laparotomy then close the abdomen.Intraperitoneally inject normal saline(0.5ml/100g)15min after the surgery.?Severe acute pancreatitis group(SAP group,n=24):use 5%sodium taurocholate(0.05ml/100g)to inject at many points beneath the pancreatic capsule,inducing SAP model.Intraperitoneally inject normal saline(0.5ml/100g)15min after the surgery.?BN52021 treating group(BN group):use the same method to induce SAP model.15 mins after inducing SAP model,intraperitoneally inject BN52021(0.5mg/100g).Choose 8 rats randomly from each group at 8h,12h and 24h after surgery to take blood in abdominal aorta and take pancreas specimens after execution.The blood used to measure plasma amylase,whole blood and plasma viscosity.they were detected by biochemical methods.plasma angiotensin ? were detected by ELISA;the pancreatic specimens were stained with HE and was made the paraffin sections.It was observed and scored under microscope.If rats died unexpected,replace the dead one by new rats.Results:1-Serum amylase:Compare the serum amylase concentration of the three group which taken from the rats respectively at 8h,12h and 24h.The result is that at 8h,12h and 24h,the serum amylase concentrations of BN group and SAP group are both higher than NC group.The differences are statistically significant(P<0.05).At 8h,12h and 24h,the serum amylase concentrations of BN group are lower than SAP group.The differences are statistically significant(P<0.05).2.Angiotensin ?:Compare the angiotensin ? concentration of the three group which taken from the rats respectively at 8h,12h and 24h.The result is that at 8h,12h and 24h,the angiotensin ? concentrations of BN group and SAP group are both higher than NC group.The differences are statistically significant(P<0.05).At 12h and 24h,the angiotensin ? concentrations of BN group are lower than SAP group.The differences are statistically significant(P<0.05).But at 8h,the angiotensin ?concentrations of BN group and SAP group are not statistically significant(P?0.05).3.Pancreatic tissue pathology score:Compare the pancreatic tissue pathology grade of the three group which taken from the rats respectively at 8h,12h and 24h.The result is that at 8h,12h and 24h,pancreatic tissue pathology score of BN group and SAP group are both higher than NC group.The differences are statistically significant(P<0.05).At 8h,12h and 24h,the pancreatic tissue pathology score of BN group are lower than SAP group.The differences are statistically significant(P<0.05).4.Whole blood viscosity:Compare the whole blood viscosity under each share rate of the three group which taken from the rats respectively at 8h,12h and 24h.The result is that at 8h,12h and 24h,the whole blood viscosity under each share rate of BN group and SAP group are both higher than NC group.The differences are statistically significant(P<0.05).At 8h,12h and 24h,the whole blood viscosity under each share rate of BN group are lower than SAP group.The differences are statistically significant(P<0.05).5.Plasma viscosity:Compare the plasma viscosity of the three group which taken from the rats respectively at 8h,12h and 24h.The result is that at 8h,12h and 24h,the plasma viscosity of SAP group is higher than NC group.The differences is statistically significant(P<0.05).The plasma viscosity of BN group is higher than NC group.But viscosity of SAP group is higher than NC group.The differences is statistically significant(P<0.05).The plasma viscosity of BN group is higher than NC group.But except at 24h,the other difference is not statistically significant(P?0.05).At 12h and 24h,the plasma viscosity of BN group are lower than SAP group.The differences are statistically significant(P<0.05).At 8h,the plasma viscosity of BN group is lower than SAP group,but the difference is not statistically significant(P?0.05).Conclusions:1.Whole blood viscosity and plasma viscosity both increase when server acute pancreatitis occurs.At the same time,angiotensin ? concentration increase too.These may be the reason for microcirculation disorder which aggravate the server acute pancreatitis.2.Ginkgolide B can correct server acute pancreatitis microcirculation disorder by reducing whole blood viscosity,plasma viscosity and angiotensin ? concentration.This may relieve or cure server acute pancreatitis.
