Objective:To observe the changes of blood viscosity and angiotensin II after using PNS to intervene rats with severe acute pancreatitis.So as to explore the therapeutic mechanism of PNS on pancreatitis and the pathogenesis of pancreatitis from microcirculation disorder theory.Method:72 healthy adult SD rats,weighting from 250g to 300g,no gender restrictions,were randomly divided into three groups:sham operation group(SO group),n=24;severe acute pancreatitis group(SAP group),n=24;panax notoginseng saponins group(PNS group),n=24.1.sham operation group(SO group):gently touched the pancreas three times after laparotomy,then close the abdomen;2.severe acute pancreatitis group(SAP group):4%sodium taurocholate(0.05ml/100g)was injected at more than one places beneath the pancreatic capsule,to inducing SAP model.3.panax notoginseng saponins group(PNS group):before inducing pancreatitis model,intraperitoneally injected PNS(50mg/ml Xuesaitong Injection(0.1ml/100g)).After injection 1H,according to the above SAP group methods to establish severe acute pancreatitis model.The blood,ascitic fluid and pancreatic specimens of rats were collected in postoperative 8h,12h and 24h.The volume of ascites was measured by watching;the plasma amylase,whole blood and plasma viscosity were detected by biochemical methods;plasma angiotensin II were detected by ELISA;the pancreatic tissue was made the paraffin sections and was stained with HE,the paraffin sections was observed and scored under microscope.Results:1.plasma amylase:plasma amylase of the three groups in postoperative 8h,12h,24h were compared,compared to the SO group,the plasma amylase concentration of SAP group and PNS group was significantly increased at all postoperative time point,the differences were all statistically significant(P<0.05);and the differences of plasma amylase concentration between SAP group and PNS group in postoperative 8h,12h,24h were not statistically significant(P>0.05).2.pancreatic tissue pathological scoring:in postoperative 8h,12h,24h pancreatic tissue pathological scoring of SAP group and PNS group was higher than the SO group,the differences were statistically significant(P<0.05);in postoperative 8h?12h?24h,pancreatic tissue pathological scoring of SAP group was higher than that of the PNS group,the differences were statistical significance(P<0.05).3.blood viscosity values:in postoperative 8h,12h,24h the whole blood viscosity values under each shear rate of SAP was higher than that of the SO group and PNS group,the differences were statistically significant(P<0.05);in postoperative 8h,24h the whole blood viscosity values under each shear rate of PNS group of was higher than that of the SO group,the differences were statistically significant(P<0.05);in postoperative 12h the whole blood viscosity values under each shear rate of PNS group of was not higher than that of the SO group,the differences were not statistically significant(P ? 0.05).In postoperative 8h,12h,24h the plasma viscosity of SAP group was higher than that of the SO group and PNS group,the differences were statistically significant(P<0.05);in postoperative 8h,24h the plasma viscosity of PNS group was higher than that of the SO group,the differences was statistically significant(P<0.05);in postoperative 12h the plasma viscosity of PNS group was not lower than that of the SO group,the differences was not statistically significant(P>0.05).4.plasma angiotensin ?(Ang ?):in postoperative 8h,12h and 24h the plasma angiotensin ? concentration of SAP group and PNS group was higher than that of the SO group.the differences were statistically significant(P<0.05);in postoperative 8h,12h and 24h the plasma angiotensin II concentration of SAP group was higher than that of the PNS group,the differences were statistically significant(P<0.05).5.Ascitic fluid:in postoperative 8h,12h and 24h the ascitic fluid volume of the SAP group and the PNS group were higher than that of SO group,the differences were statistically significant(P<0.05);in postoperative 8h,12h and 24h the ascitic fluid volume of the SAP group were higher than that of PNS group,the differences were statistically significant(P<0.05).Conclusion:1.when severe acute pancreatitis occurs,the whole blood viscosity,plasma viscosity and the plasma angiotensin II concentration increase,which may lead to microcirculation disturbance in pancreatic tissue.2.Panax notoginseng saponins may be able to reduce the blood viscosity and plasma angiotensin II concentration to alleviate microcirculation disturbance,so as to achieve the role of curing severe acute pancreatitis.
