Background: the cardiovascular disease has become a problem of global health care.(Coronary Heart Disease(CHD),also known as coronary arteriosclerotic heart disease, refers to coronary artery atherosclerosis or spasm caused by vascular stenosis or obstruction, coronary circulation disorder, resulting in myocardial ischemia, hypoxia or necrosis caused by heart disease.Triglyceride(TG)metabolic disorder is an independent risk of hypertriglyceridemia. The factors of Coronary Heart Disease(CHD).The discovery of apolipoprotein C3(APOC3)gene is associated with an increase in the level of TG.The aim of this study was to investigate the risk of coronary heart disease, blood lipid and blood pressure in Chinese people with two gene(rs4520 rs5128)polymorphisms.Methods: a total of 381 patients who were admitted to Qingdao municipal hospital in December 2016 December and underwent coronary angiography or coronary artery CT angiography were selected in December. The results of coronary angiography and medical history were divided into groups: 229 cases, control group: 152 cases, application of MassArray genotyping method for studies of rs4520 with rs5128 for determination of gene polymorphism, determination of blood lipid levels, and blood pressure, diabetes and smoking history, according to the results of correlation analysis between APOC3 gene rs4520 and rs5128 polymorphism and coronary heart disease.Results:1.The two independent samplest test was used to compare the numerical variables between the groups.chi square test was used to compare the categorical variables between the two groups. The results showed that there was no significant difference between BMI(P=0.637).Obesity(P=0.281)and Hypertension(P=0.098)in the distribution of coronary heart disease and non CHD group.Age(P=0.023),Total cholesterol(P<0.001),Triglyceride(P<0.001),High density lipoprotein(P<0.001),Low density lipoprotein(P<0.001),Gender(P=0.01),Diabetes mellitus(P<0.05),Smoking(P<0.05)in the difference between the two groups was statistically significant.According to the mean, the age, total cholesterol and low density lipoprotein in the case group were higher than those in the control group. In addition, the percentage of men,the proportion of diabetes, and the proportion of smokers in the case group were higher than those in the control group.2.The APOC3 rs4520 gene has no statistical significance in the risk of CHD(P>0.05).The APOC3 rs5128 genotype has no statistical significance in the risk of CHD(P>0.05).3.A single factor analysis of variance was used to compare the differences among the various genotypes in the each genotype of APOC3 rs4520 and blood lipid between the case group with the control group. In the case group of the APOC3 rs4520 gene, the difference in total cholesterol levels between the three genotypes was statistically significant(P<0.05),and the highest in the TC group. Moreover, from the frequency of APOC3 rs4520 TC allele, compared with triglyceride,high-density lipoprotein and low density lipoprotein group, there is an increasing trend relative to CC and TT alleles.4.A single factor analysis of variance was used to compare the differences among the various genotypes in the each genotype ofAPOC3 Rs4520 and blood lipid between the case group with the control group. In the case group and the control group of the APOC3 rs5128 gene, there was no significant difference in the difference of blood lipid among the three genotypes. But from the gene frequency of APOC3 CG alleles, there is a trend of increasing relative to CC and CG alleles in triglyceride,high-density lipoprotein and low density lipoprotein group.5.In the analysis of Rs4520 in the group of hypertension and diabetes, using single factor Logistic regression analysis, the prevalence of diabetes and hypertension, were as dependent variable, with three Rs4520 genotypes as independent variables, the results showed that: 1.in the hypertension group, compare with the wild-type CC genotype,TC genotype proportion were higher in hypertensive patients, with statistical difference(P<0.05),the TC genotype increased risk of hypertension(OR=2.206,95%CI:1.193-4.028,P=0.012<0.05).2.,in diabetic group, there was no significant difference between TT and TC alleles compared with CC genotype(P>0.05).APOC3 Rs4520 allele did not increase the risk of diabetes.6.In the analysis of Rs5128 in the group of hypertension and diabetes, using single factor Logistic regression analysis, the prevalence of diabetes and hypertension, were as dependent variable, with three Rs5128 genotypes as independent variables, the results showed that: 1.in the hypertension group, each genotype ofAPOC3 Rs5128 has not statistically significant in the risk of the hypertension(P>0.05),Rs5128 did not increase the risk of hypertension.2.in the diabetic group, each genotype ofAPOC3 Rs5128 has not statistically significant in the risk of the diabetic(P>0.05),Rs5128 did not increase the risk of diabetes.Conclusion:1.The polymorphism of APOC3 rs4520 gene can not increase the risk of coronary heart disease in the population of Qingdao.However,APOC3 rs4520 TC allele increases the total cholesterol level and hypertension,it needs to be further confirmed.2.The polymorphism of APOC3 rs4520 gene can not increase the risk of coronary heart disease in Qingdao.
