Effects Of Apolipoprotein A5-1131T>C Polymorphism On Blood Lipids And Coronary Heart Disease And Strokes | Posted on:2007-02-28 | Degree:Doctor | Type:Dissertation | Country:China | Candidate:X P Li | Full Text:PDF | GTID:1104360185986763 | Subject:Department of Cardiology | Abstract/Summary: | PDF Full Text Request | The effect of the single mucleotide polymorphism -1131T>C in the apolipoprotein A5 gene on blood lipidsBackground: A newly discribed member of the apolipoprotein gene family, apolipoprotein A5 (APOA5), was discovered by comparative sequencing between human and mouse APOA1/C3/A4 gene cluster in 2001. The APOA5 gene has been showed to play an impotant role in determing the metabolism of blood lipids especially triglyceride(TG) in animal models and humans. The studies of common single mucleotide polymorphisms (SNPs) of APOA5 in different ethnic groups indicated that the carriers of rare alleles had higher TG levels. There is an ethnic difference in APOA5 gene variants. Objective: To investigate the gene frequencies of APOA5 -1131T>C variants and the association between the -1131T>C polymorphism and blood lipids in a Chinese Han population.Method: Polymerase chain reaction-restricted fragments length polymorphism (PCR-RFLP) and polyacrylamide gel electrophoresis (PAGE) methods were used to determine the APOA5 -1131T>C genotypes in 247 healthy individuals. The levels of serum lipid profiles were also analyzed by enzymatic methods.Results:1. The APOA5 -1131T>C genotype distribution in healthy individuals of...
