Investigation On That Anti-inflammatory Mechanism Of Fire Needle Prick In The Acute Gouty Arthritis Rat Model With Syndrome Of Accumulated Dampness-heat

Objective:To acute gouty arthritis rat model with syndrome of accumulated dampness-heat as experimental object,explore the fire needle prick anti-inflammatory mechanism of treatment of acute gouty arthritis,provide experimental basis for clinical application.Methods:The 32 SD rats were randomly divided into 4 groups,8 in each group,namely blank group,model group,fire needle group and western medicine group.In addition to the normal feeding of the blank group,the other three groups were all under the high-sugar and high-fat diet for 10 days,and the hot and humid environment lasted for 5 days,the syndrome of accumulated dampness-heat model was built.In the 24 hours after the model rat model was completed,the model of Acute gouty arthritis(AGA)was established by injecting 0.1ml of sodium urate into the right ankle joint of rats.After 24 hours,the model group was bound for 10min/day.The highest point of the right ankle swelling of the right ankle of the fire needle is the"ah is the point",,one time/day;The western medicine group gave colchicine 0.1mg/100g to the stomach,one time/day;Each group was treated for 3 consecutive days.To observe the general state after the treatment of the rat model before and after the treatment.The right foot gait of 24hAGA rats was observed after Oh,24h after molding and 24hAGA after treatment.The swelling degree of the right ankle joint was observed after 4,8,12,24 and after the treatment.The content of il-1,il-6 and tnf-alpha in the synovial tissues of rat’s ankle was determined by ELISA.Results:1.Began to build the syndrome of accumulated dampness-heat model,the rat burnout compulsive lying lazy action dull,thin and fluffy hair color,reduced the volume of food,drinking water,yellow urine,stool again pond have diarrhoea or gooey,the last part of the rats appear heavy breathing,dysphoria,hair loose coarse,defecate pond diarrhea or gooey symptoms aggravate,gradually see crissum filth;Consistent with syndrome of accumulated dampness-heat.Fire needle prick bloodletting,colchicine treatment rats after burnout,compulsive lying lazy move,action dul,the hair fleeciness was improved,food,drinking water quantity was increased,the yellow color of urine,defecate pond diarrhea rats or gooey deformed.2.Groups of rats after acute gouty arthritis building 24 hours gait score results:24 hours after modeling:except for the blank group,the difference between the model group,the fire needle group and the western medicine group was statistically significant(P<0.01).After the treatment,24h:the model group,the fire acupuncture group and the western medicine group were significantly decreased after 24h after the molding,and the difference was statistically significant(P<0.01).After 24h and 24h after the treatment,the gait scores were increased and the difference was statistically significant(P<0.01).There was no statistically significant difference between model group,fire needle group and western medicine group(P>0.05).3.Groups of rats right ankle swelling degrees of different time test results:Each group was made before and after comparison:Compared with the pre-mold Oh,after the blank group,the swelling degree of the right ankle of the rats in the treatment group was increased by 12h and 24h,and the difference was statistically significant(P<0.01).Each group was compared before and after treatment:For 24h after treatment the treatment group rats right ankle swelling degree decline,including model group difference was statistically significant(P<0.01),fire needle group and the western medicine group differences have statistical significance(P>0.05);Compared with 12h and 24h after mold building,the swelling degree of each treatment group decreased at 24h after treatment,and the difference was statistically significant(P<0.05,P<0.01).12h and 24h after mold making:the comparison between each treatment group and the blank group increased,and the difference was statistically significant(P<0.01).The difference between the model group and the blank group was statistically significant(P<0.01),and the difference was statistically significant(P<0.05).There was no statistically significant difference between the acupuncture group and the western medicine group(P>0.05).4.Results of IL-1β,IL-6 and TNF-α:compared with the blank group,there was a significant increase in IL-1β,IL-6 and TNF-α in the synovial tissues of the model group,and the difference was statistically significant(P<0.05,P<0.01).Compared with the model group,the contents of IL-1β,IL-6 and TNF-α were significantly decreased in the synovial tissues of the fire needle group and the western medicine group,and the difference was statistically significant(P<0.05,P<0.01).There was no significant difference between the fire acupuncture group and the western medicine group,and the difference was not statistically significant(P>0.05).Conclusion:1.The fire needle prick and the western medicine colchicine can improve the gait and the swelling degree of the ankle joint,reduce the content of IL-1β,IL-6 and TNF-α in the synovial tissues of acute gout arthritis,and the efficacy is comparable.2.The anti-inflammatory mechanism of the puncture of The fire needle prick in the treatment of acute gouty arthritis may be related to the inhibition of IL-1β,IL-6 and TNF-α in the synovial membrane.
