Objective:The experimental observation by acupoint Jieyufang intervention change behavior of depression model rats,and the changes of Raf1 and p-ERK in the frontal cortex and hippocampus of depressed rats,to further study the mechanism of the treatment of depression in the ERK signaling pathway,to provide more scientific and reliable mechanism based for the clinical acupoints Jieyufang on the intervention of depression.Methods : 30 healthy male SD rats were randomly divided into 3 groups: blank control group(n=10),model group(n=10)and acupuncture group(n=10),1 day before the beginning of the experiment,3 groups of rats underwent open field test,sugar experiment,weight,recording performance of these behaviors.during the experiment,Blank control group of normal feeding,not to mold the stimulation;Model group and acupuncture group were treated with single cage feeding combined with unpredictable mild stimulation to prepare rat depression model,stimulated source selected a total of 7 species,daily random selection of a way,a total of 21 consecutive days to accept the different stress modeling.Group on the daily before modeling of 1H acupuncture acupoints Jieyufang(Baihui +Shenmen + Taichong),one times a day,20 minutes each time,a continuous electroacupuncture treatment time is 21 days,crawling and fixed model group,no acupuncture operation,the other treatments were the same as those in the acupuncture group,the blank control group did not give any treatment.After the experiment the rats again for the open field test,sugar experiment,weighing behavioral evaluation and record,the rats were decapitated after anesthesia,rapidly dissected rat hippocampus and prefrontal cortex,-80 centigrade refrigerator to be measured.The method of enzyme-linked immunosorbent assay was used to test the changes of Raf1 and p-ERK contents in the prefrontal cortex and hippocampus of rats.Results:Score 1: behavior before the start of the experiment,3 groups of rats in the open field test and sugarexperimen and body weight behavior were no statistically significant difference(P > 0.05).After the end of the experiment,in the open field test scores of horizontal and vertical erection times,sugar experiment,body weight of rats,the model group was compared with the blank control group,score data were significantly decreased,there was a significant difference(P <0.01),suggesting that the preparation ofthis depression model was successful.In the open field test,sugar experiment,acupuncture group compared with model group,score increased significantly,there was a significant difference(P <0.01),In terms of body weight in rats,acupuncture group and model group were compared,the result is increased,the difference was statistically significant(P<0.05),suggesting that electroacupuncture acupoints Jieyu Fang intervention in depression of rats in all aspects of its behavior has improved and the effect of treatment.2.Changes of Raf1 and p-ERK in brain?Hippocampus: contrast with the blank control group,the contents of Raf1 and p-ERK in the hippocampus of the model group were significantly decreased(p <0.01),there was a statistically significant difference.Compared with the model group,the contents of Raf1 and p-ERK in the hippocampus of the acupuncture group were significantly increased(P<0.01),differences were statistically significant.Frontal cortex:the levels of Raf1 and p-ERK in the model group were significantly lower than those in the blank control group(P <0.01),the difference was statistically significant;the acupuncture group and the model group for comparison,the contents of Raf1 and p-ERK were significantly increased(P<0.01),there is a significant difference in comparison.Conclusion:1.The combination of single cage feeding and chronic unpredictable mild stimuli can have a reduced effect on the behavioral aspects of rat,so that rats had depressive symptoms.Acupuncture acupoints Jieyufang of the intervention can improve the performance of depression in the performance of rats..2.The contents of Raf1 and p-ERK in the frontal cortex and hippocampus of depressed rats were decreased,suggesting that depression occurs with the reduction in the ERK signaling pathway,acupoints Jieyufang intervention can improve the levels of Raf1 and p-ERK in the brain of depressed rats,may activate the expression of the ERK signaling pathway,this may be the acupoints Jieyufang treatment of depression in the ERK signaling pathways play a mechanism.
