Objective:Employ the transgenic mice expressing psoriasis K5.STAT3C were selected as subjects.Through the experimental study of centipede antiphologistic decoction detection of transgenic mouse model of IL-23 in peripheral blood and skin lesions and the correlation effect,at the same time,the correlation between IL-23 and skin lesions was analyzed..Methods:1.Model:K5.STAT3C transgenic psoriasis mice were randomly divided into 5 groups,each group has 5,they were centipede antiphologistic decoction low-dose group,centipede antiphologistic decoction dose group,centipede antiphologistic decoction high dose group,methotrexate western medicine control group and normal saline blank control group.5 groups of transgenic mice were given 2ml/100g/d daily,two times a day.The control group of each dose group and the saline blank control group were filled with 8d continuously;the methotrexate western medicine control group was given a continuous gavage of 4d,after that,4d was gavage with saline,the dose and frequency of gavage are the same as before.This method is suitable for methotrexate in clinical patients.1h after last administration,the transgenic mice were anesthetized by intraperitoneal injection of pentobarbital sodium(35mg/kg).Fix the mice on the operating table,blood extraction from the abdominal aorta under the aseptic condition 4ml.The extracted blood was mixed evenly in heparin sodium and vacuum blood collection,to room temperature preservation.And PBMC was extracted within 2 hours for cytological detection.2.Group intervention:1)To count the MPSAI score of skin lesions in m ice;(2)Detection of IL-23 level in mice serum by ELISA;(3)The correlati on between IL-23 and skin lesions was analyzed.Results:1.MPSAI score of skin lesions:Methotrexate western medicine control group,the MPASI score in the high dose group of centipede sepsis drink was the biggest,that is,the highest MPASI value.Low dose of centipede in the middle dose group and low dose group were lower;the smallest change in the normal saline control group,the lowest Delta MPASI value.2.IL-23 content:Comparison with blank control group,the content of serum IL-23 factor had statistically significant differences in the centipede antiphologistic decoction dosage group(P<0.05);Control group,centipede antiphologistic decoction high dose group had significant difference in western medicine methotrexate(P<0.01);at the same time,the content of IL-23,blank control group and low dose group of centipede antiphologistic decoction compared,the difference was statistically significant(P<0.05).Compared with the control group of Western medicine,centipede antiphologistic decoction,in low dose group were statistically significant(P<0.05).While the control group of Western medicine and centipede antiphologistic decoction high dose group compared with no significant difference(P>0.05).The serological content of serum IL-23 factor is explained,the centipede antiphologistic decoction in the high dose group and the methotrexate group had lower serum levels of function,and the concentration of centipede antiphologistic decoction higher serum content is low,the inhibitory effect is more obvious,inhibitory effect of methotrexate and Western medicine centipede antiphologistic decoction high dose group were similar.3.The level of IL-23 serum factor and the change of skin lesion in transgenic mice before and after administration.The delta MPASI variable linear regression,the corresponding correlation coefficient is calculated by this line,Its p=0.000<0.05,to explain the establishment of a straight line,there is correlation between IL-23 and MPASI.At the same time,correlation coefficient is negative,so correlation is negative correlation.The mice to delta MPASI before and after administration of the larger,it indicates that the higher the remission degree of skin lesions is,the more effective the treatment is.The lower the serum IL-23 is.Conversely,the smaller the delta MPASI value,the lower the remission degree of skin lesions and the less effective treatment.The higher the IL-23 value in serum is.This is consistent with the expression function of IL-23 and the correlation function between psoriasis and literature.Conclusions:1.Centipede antiphologistic decoction can reduce the MPASI score and improve the clinical symptoms of transgenic mice.2.Centipede antiphologistic decoction can reduce serum IL-23 level in transgenic mice.3.The level of IL-23 serum factor was correlated with the change of skin lesions before and after administration in transgenic mice,and it is negative correlation4.Centipede antiphologistic decoction can be used as an effective way to treat psoriasis in clinic,and provide a theoretical basis for its treatment.
