Objective:To observe the protective effect of rhubarb on acute liver injury in mice induced by intragastric administration of high-dose centipede decoction.Methods: Sixty KM mice were randomly divided into five groups namely,normal control group,centipede liver injury group,rhubarb low-dose treatment group,rhubarb medium-dose treatment group,and rhubarb high-dose treatment group,with12 mice in each group.Adaptive breeding in mice after 1 w,according to the group were given 0.4 ml drug lavage,in addition to the control group,both the decoction lavage centipede fluid is 6.5 g/kg content(clinical dose equivalent to 10 times),and rhubarb low,medium and high dose treatment group respectively lavage with rhubarb decoction fluid is 0.048 g/kg content,content of 0.0975 g/kg,0.195 g/kg content(equivalent to 1/8,1/4,1/2 of clinical dose),28 d continuously.The general condition,activity state,food intake,defecation and body weight of mice were observed.After28 days,the serum levels of Alanine aminotransferase(ALT),Aspartate aminotransferase(AST),Alkaline phosphatase(ALP)and Total Bilirubin(TBil)were detected,and the specific gravity of the liver was measured.The pathological changes of the liver of each group of mice were observed by HE staining.At the same time,Western Blot was used to determine the expression of quinone oxidoreductase1(NAD(P)H:quinone oxidoreductase1(NQO1)and hemeoxygenase-1(ho-1)proteins in mouse liver tissues.Results:1.Compared with the normal control group,ALT,AST,ALP and TBil contents in serum of mice in the centipede liver injury group were significantly increased,with statistically significant differences(P<0.05);ALT,AST,ALP and TBil contents in serum of mice in the rhubarb high-dose treatment group were also significantly increased,with statistically significant differences(P<0.05).Compared with the centipede liver injury group,the content of ALT,AST,ALP and TBil in the serum of the rhubarb low-dose treatment group was significantly decreased,and the difference was statistically significant(P<0.05).The AST of the rhubarb medium-dose treatment group was significantly decreased,and the difference wasstatistically significant(P<0.05).2.The high-dose centipede decoction can lead to the occurrence of liver injury and liver pathological changes in mice after being gavaged for 28 days,and the improvement was most obvious in the low-dose rhubarb group.3.Compared with normal control group,in Nrf2 / ARE pathways NQO1,HO-1protein expression significantly enhanced in the centipede liver injury group,the difference was statistically significant(P<0.05),rhubarb high,medium and low dose treatment group NQO1,HO-1 enhancement,the centipede liver injury group of protein expression difference was statistically significant(P<0.05,P<0.01),with rhubarb enhanced low dose group is significant(P<0.01).Conclusion:1.High-dose centipede water decoction can cause mild acute liver injury in mice after 28 days of intragastric administration.2.Low and medium doses of rhubarb can alleviate acute liver injury caused by intragastric administration of high-dose water decoction of centipede.The effect of low-dose rhubarb is the best.3.The effect of rhubarb "promoting aging and causing new effects" can regulate the level of oxidative stress in the liver by increasing the expression of NQO1 and ho-1proteins,so as to reduce the acute liver injury in mice caused by high-dose centipede decoction.
