The Wohlfahrtia magnifica mainly causes the female camel vaginal maggot disease,is one of the main diseases that affect the healthy development of the bactrian camel,and brings serious economic losses to the camel breeding industry.The disease occurs from May to October each year,and the pathogen resolves spontaneously in the winter,and relapses again in May of the following year.The purpose of this experiment is to reveal the wintering behavior of the Wohlfahrtia magnifica,the survival rate at low temperature,and the low-temperature transcriptome study.The research has potential significance for controlling pests and guaranteeing production and life in pastoral areas.1?The Wohlfahrtia magnifica flies through the winter in the form of pupae.2?With the decrease of temperature,the feathering rate of pupae also decreases,but the feathering rate is similar at-5?,-10?,and 4? and is 48.75%,47.50%,and 50%respectively.At-15?,the feathering rate decreased by a cliff and the feathering rate was 3.75%.3?Through comparative analysis of transcriptome sequencing,NCBI_nr,GO,KEGG,Pfam,Swiss-Prot and eggNOG database comparisons were annotated to 39197 unigenes,of which PA1vsPA was increased by 13465 unigenes,25648 unigenes were down,no84 unigenes were down,PA2vsPA was up 14572 unigenes,down 24578 unigenes,no47 unigenes.Through differential gene analysis,the HSPs family genes Hsp23,Hsp27,Hsp67bc,Hsp70Ba,Hsphl,Hsp60,Hsp83,Hsp90AB1/AA1(log2FC>2,p<0.05)and Hsp homologous protein Hsc gene Hsc70 5.Hsc70-4(log2FC>2,p<0.05).Five genes Hsp67bc,Hsp23,Hsp27,Hsc70-4,Hsp70Ba with higher expression levels were selected for q-PCR verification.The difference in Hsp27 expression was extremely significant(p=0.009<0.01);the difference in Hsp23 was significant(p=0.037<0.05);Hsc70-4 was significantly different(p=0.026<0.05);Hsp70Ba was significantly different(p=0.039<0.05);Hsp67Bc was not significantly different(p=0.271>0.05).This experiment supplements the research on the life behavior of the Wohlfahrtia magnifica and provides a basis for later research.A transcriptome database at different temperatures of the pupa of the Wohlfahrtia magnifica was established,which revealed the expression changes of related genes in the pupa of the Wohlfahrtia magnifica at low temperature,providing a basis for further research in the future.
