Wohlfahrtia magnifica is the main pathogen of fly wound myiasis in many countries in Europe,Africa and Asia. The life history of Wohlfahrtia magnifica is affected by the season and cannot meet people’s needs for sampling of pathogens at any time.In this thesis, the artificial feeding of Wohlfahrtia magnifica is studied,so that the metamorphosis life history of Wohlfahrtia magnifica is not affected by seasonal changes, and to ensure that Wohlfahrtia magnifica, pupae and larvae reproduce all year round. It provides convenient conditions for studying the prevention and control of myiasis caused by Wohlfahrtia magnifica. The results of study are as follows:1.Artificial cultivation of Wohlfahrtia magnifica larvae:The pupation rate of the third generation larvae cultured in vitro was about 84%,and it took 3-4 days for the first stage larvae to mature the third stage larvae.2.The pupae were cultured artificially(1)Compared with F0 larvae, the weight of F1 and F2 larvae transformed into pupae increased by 13.3 mg.(2)The emergence rate of F1 and F2 pupae was 40.33-43.33% higher than that of F0 pupae.3.Artificial preservation of pupae:The pupae could be stored for 30-60 days at 4 ℃.4.Artificial cultivation of Wohlfahrtia magnifica:(1)Female and male identification: With the help of dissecting microscope, the abdomen of adult male flies is long oval, the abdomen of female adult flies is oval, and the abdomen of male flies is narrower than female flies. The tail organs of males and females are the most distinct. The end of the abdomen of males is smooth, the tail organs are convex, and the dorsal plate of the end of the abdominal segment forms a sac-shaped protrusion with a dark yellow bulge and oblique downwards, showing a spoon-shaped swelling;grayish white.(2)The longest lifespan of the captive Wohlfahrtia magnifica is 62 days, with an average of 36.18 days.(3)The average number of maggots produced by artificially reared female flies is45.(4)Sexual maturity and mating appeared on the 4-5th day after eclosion, and copulation was more common in the daytime. The mating peak was entered on the 3rd day,and the mating period began to decrease slowly on the 10 th day.The shortest mating time is about 2h and the longest is about 4h. |
