This experiment mainly studies the haploid technology of sweet corn.The experiment content is divided into three technical links: factors affecting haploid induction,sweet corn haploid multiplication and drug selection,and the related combining ability analysis of sweet corn DH line.The results of the test are as follows:(1)Sweet corn haploid induction test: The test mainly from different length of filaments,inducing system of loose powder time,different genotypes of inducible lines,different genotypes of sweet corn hybrids,four experimental steps to carry out the study of induction rate,filament length The average induction rate was between 8.26% and 4-8cm,which was significantly higher than the average induction rate between 0-4cm and 5.79%.This indicated that sweet corn was the most suitable for induction when the filament length was 4-8cm.The average induction rate of inducing system loose powder at 0-5t was 5.78%,which was significantly lower than the mean induction rate of loose powder at 5-10 t,which indicated that the time of application of the powder for the delayed induction system could increase the induction efficiency because the induction system was effective.Pollen viability and pollen amount are best in the loose powder for 5-10 days.The inducing rates of different inducing lines and different genotypes of sweet corn hybrids reached extremely significant differences,indicating that the difference in the inducing material’s blood was an important factor affecting the haploid induction rate of sweet corn.(2)Sweet corn doubled haploid test: using 40-80μmol/L trifluralin,bispyribacillin,and acetochlor soaking seeds to double the sweet corn haplotype,the results showed that trifluralin with 80μmol/L The best treatment of Ji Tian 99,the maximum rate of up to 12.34%.Extremely significant or significant differences were observed between herbicides of different concentrations.The doubling method used soaking,immersing,dipping,acupuncture growing points,and dripping leaves,naturally doubling,and doubling the dosage as a mixed solution of colchicine and DMSO.The analysis of variance showed that the doubling method reached extremely significant differences.Among them,the doubling efficiency of the budding method is the best,with an average of 10.95%.Therefore,the doubling method is an important factor affecting the doubling effect of sweet corn haplotypes.The effects of soaking time and colchicine concentration on the haploid doubling effect of sweet corn were further studied under the same constant temperature of 20°C.The results showed that the average doubling rate was up to 6.11% at the soaking time of 12 hours,which was different from other processing time.Significant.The average doubling rate of colchicine concentration of 0.6 mg/mL was as high as 8.49%,which was significantly different from the concentration of other agents.Therefore,at the constant temperature of 20°C,the doubling rate of sweet corn haplotypes treated with 0.6 mg/mL colchicine solution was the highest.(3)Heterosis analysis of DH lines: Based on the analysis of average dominance and super-parent heterosis,the results showed that the average dominance value of single panicle weight was up to 167.32%,and the value of super-parent advantage was the highest,reaching 234.15%.According to the heterosis index,the average value of the traits of the cross combinations was: plot yield> single ear weight> ear position> 100 kernel weight> plant height> number of ear rows> number of row grains> soluble total sugar> ear coarseness > Spike length> stem diameter> baldness length> rind rate.(4)Analysis of combining ability: Analysis of general combining ability and specific combining ability of the sweet corn hybrid parents.The general combining ability of the DH line indicates that the number of soluble sugars of the DH line No.N5 is the largest,so the heritability of the sugar-containing gene is the highest.Can be used as a high sugar content hybrid parent material.Therefore,the best DH lines N5,N4 and N1 can be selected as the ideal sweet corn breeding inbred line by the general combining force value.Specific combining ability analysis was performed on 30 hybrid combinations.The results showed that the combinations with the best comprehensive traits included N16,N17,N22,and N24.These combinations included plant height,ear position,stem diameter,panicle weight,and 100-grain weight.The agronomic traits all had a number of higher specific mating force values,of which the largest combination of the total specific sugars of soluble sugars was 41 and reached 26.7942.
