Objective:In this study,early warning of cows with metritis was carried out,and Jiangongsan was used to treat cows with metritis.The blood metabolism,hormones and morphological indexes of cows before and after treatment were measured.To explore the effect of Jiangong Powder on blood metabolism,hormones and morphology of cows with metritis.The aim is to provide a basis for better monitoring and prevention of metritis in the future,to provide certain technical support for the prevention and treatment of metritis,and to provide a scientific basis for clinical application of the drug.Methods:(1)The levels of E2,P4,FSH,PGE2 and AST in the serum of cows in normal and diseased groups were detected by enzyme-linked immunosorbent assay(ELISA).Pearson correlation analysis,binary logistic regression model and ROC analysis were used to obtain early warning indicators and determine their threshold.(2)The Jiangong Powder was administered by the irrigation device,and the number of red blood cells,hemoglobin,lymphocytes,white blood cells and granulocytes in the blood of the normal group,the diseased group and the treatment group before and after treatment were measured by automatic blood cell analyzer.The levels of BHBA,BUN and AST in the serum of the normal group,the diseased group and the treatment group before and after treatment were measured by ELISA,and statistical software was used for analysis.(3)The contents of E2,P4,FSH,LH,PGF2αand PGE2 in serum of each group were detected by ELISA and analyzed by statistical software.(4)B-ultrasound was used to monitor the morphological parameters of the uterus of the normal group,the diseased group and the treatment group at 742 days postpartum.Results:(1)The results showed that 3 indicators of AST,P4 and PGE2 at 59 days postpartum could be used as an early warning indicator for cows with metritis.AST>6.39 ng·mL-1,P4>9.53 ng·mL-1,PGE2>3.92ng·mL-1,there was a risk of having mastitis in postpartum.(2)The results showed that the number of red blood cells,hemoglobin,lymphocyte ratio,BUN and AST in the normal group before treatment were significantly higher than those in the diseased group and the treatment group(P<0.05);the number of white blood cells,granulocytes and BHBA were significantly lower than the disease group and the treatment group(P<0.05);the content of each index in the blood after treatment was close to the normal group,and the difference was not significant(P>0.05).(3)The results showed that the levels of E2,P4,PGF2αand PGE2 in the normal group were significantly different from those in the treatment group and the diseased group(P<0.05).The level of LH in the normal group was significantly different from that in the treatment group(P<0.05),but not significantly different from that in the diseased group(P>0.05).The difference of FSH level between the groups was not significant(P>0.05).The levels of E2,FSH and PGF2αwere significantly higher than those in the diseased group(P<0.05).The levels of P4 and PGE2 were significantly lower than those in the diseased group(P<0.05).There was no significant difference between the LH level and the diseased group and the normal group(P>0.05).(4)The results showed that the uterus morphology of uterus cows was gradually restored to normal levels after treatment with Jiangong Powder.At 742 days postpartum,the cervix longitudinal diameter of the normal group,the diseased group and the treatment group showed a downward trend.At 42 days postpartum,the average cervix longitudinal diameter of the normal group,the diseased group and the treatment group recovered to 2.72 cm,2.87 cm and 2.67 cm.The cervix longitudinal diameter in the diseased group significantly higher than those in the diseased group(P<0.05),the difference between other time points was not significant(P>0.05).In the normal group,the diseased group and the treatment group,the thickness of the uterus in the uterine horn was decreased at2142 days postpartum.At 42 days postpartum,the average uterine horn wall thickness of the normal group,the diseased group and the treatment group recovered to 3.07 cm,3.36 cm,and 3.14 cm;The treatment group uterine horn wall thickness at each time point with no significant difference were observed in the disease group and the normal group(P>0.05).Conclusion:AST,P4 and PGE2 can be used as early warning indicators to predict the risk of metritis.Jiangong Powder has a regulating effect on some blood indexes,metabolic indexes and reproductive hormone levels of cows with uterus,which can improve the body’s anti-inflammatory and immune ability,improve metabolic function,promote uterine morphology recovery,and better prevent and treat metritis. |
