| Object:The aim of this experiment was to establish early warning indicators of dairy cows with uteritis,and using Jiangong Powder to treat them.At the same time,detecting the changes of some immune and antioxidant indicators in serum of dairy cows before and after treatment,in order to explore the effects of Jiangong Powder on immune and antioxidant indicators of dairy cows with uteritis,and provided theory for the prevention and control of uteritis and the treatment of dairy cow uteritis with traditional Chinese medicine.Ultimately,enhance the ability of Chinese herbal medicine to serve the modern dairy industry.Methods:?1?Using ELISA to detect the content of IL-1,IL-2,IL-8,TNF-?,IFN-?,Ig G and Ig M of serum and detecting the activity of SOD,GSH-Px and the content of NO and MDA in serum by WST-1,dithiodinitrobenzoic acid method,nitrate reductase and thiobarbituric acid?TBA?method.Utilizing Pearson correlation analysis and binary logistic regression model to obtain early warning indexes,and ROC analysis to determine the diagnostic effect and boundary value.?2?By oral Jiangong Powder to detect the contents of some immune indexes?IL-1?IL-2?IL-8?TNF-??IFN-??Ig G and Ig M?and antioxidant indexes?NO?MDA?SOD?GSH-Px?in serum before and after treatment,and use statistical software to analyse the changes of them.?3??1?Using color Doppler ultrasonography to diagnose pregnancy and uteritis in dairy cows,and obtained sonograms and descriptions.?2?Counting the effective percentage rate and recovery rate of Jiangong Powder and its combination with chloroprostenol in the treatment of dairy cow with uteritis,and by chi-square test to evaluate the treatment effect of the two schemes.Results:?1?The results showed that NO,IL-2 and IFN-?could be used as early warning indicators of dairy cow with uteritis.When NO>14.018?mol·L-1,IL-2>119.509 pg·mL-1 and IFN-?>149.563pg·mL-1,dairy cows were at risk of postpartum uteritis.?2?The result was showed that the levels of IL-1,IL-2,TNF-?and IFN-?in serum of dairy cows with uteritis before treatment were significantly higher than those of the normal group?P<0.05?,and the levels of Ig G and Ig M were significantly lower than those of the normal group?P<0.05?.After treatment,the levels of serum were close to normal,and there was no significant difference?P>0.05?,but the levels of IL-8 were not significant?P>0.05?.The content of NO in serum of dairy cows with uteritis before treatment was significantly higher than that of normal group?P<0.01?.After treatment,the content of NO in serum was significantly lower than that of normal group?P>0.05?.There was no significant difference in the content of MDA between the two groups before and after treatment?P>0.05?.There was no significant difference in serum SOD and GSH-Px activity between the two groups before treatment?P>0.05?.After treatment,the activities of SOD and GSH-Px increased gradually,but there was no significant difference between the two groups.?3?Results are:?1?The diagnosis of color Doppler ultrasonography showed that the endometrial hyperplasia was increased and the echo intensity increases in cows with uteritis.In normal pregnant cows,one or more large oval or round cotyledons could be seen in liquid dark areas,and in some dark areas,long and bright fetal membrane lines could be seen.?2?The effective rates of Jiangong Powder and Jiangong Powder plus chloroprostenol in the treatment of dairy cow uteritis were 70.79%and 85.19%respectively,and the cure rates were 51.61%and55.56%respectively,Chi-square test showed that there was no significant difference between Jiangong Powder and Jiangong Powder plus chloroprostenol of curative effect in the treatment of dairy cow uteritis?P>0.05?.Conclusion:Serum NO,IL-2 and IFN-?can be used as early warning indexes of dairy cow uteritis.Jiangong Powder can regulate some immune and antioxidant indexes in serum of dairy cows with uteritis.By improving the degree of inflammation,it can promote postpartum estrus conception and uterine involution of dairy cows,and has a good therapeutic effect on postpartum uteritis of dairy cows. |
