| In recent years,as one of prior utilized plants for biological fuel oil and energy substitute,Swida walteri(Wanger.)Sojak oil value has been gradually developed and utilized.The traditional seed reproduction or asexual reproduction is difficult to meet the needs of the short term to provide a large number of seedlings,due to the limited superior individual plant materials,so a rapid tissue culture system is necessary.The current research on S.walteri only to primary culture,with the problem of high pollution rate and low induction rate,so it can not be applied in production.This experiment was conducted to investigate in vitro micropropagation of S.walteri with the apical buds excised from the growing shoots of annual branches.(1)The best disinfection method was to use the tender shoots as explants for tissue culture,first with 75%alcohol disinfection about 30s,and then using 2%sodium hypochlorite for 15 min,contamination rate was 4.4%;(2)The optimum basic culture mediLnn for initial culture was DKW.the plantlets grew well and the leaves were healthy;(3)Use thick++stem tip(Internode length is short,in addition to an axillary bud with fist bud.the length is 4-7mm)as generational culture materials,germination rate was up to 94%;Under the condition of light intensity 2000-30001x,it is beneficial to the growth of plantlets;Thelnost suitable medium pH of S.walteri tissue culture was 6.2;To cultivate high strong plantlets,can add 1mg/L GA.,40 days after the leight of plantlets can reach 8.8cm;The best generational culture inedium was DKW witli 6-BA 0.5mg/L,NAA 0.2mg/L and GA3 0.5mg/L,the average multiplication coefficient was 15.56.average seedling height was 6.22cm.plantlets were healthy and strong;(4)The effects of different concentrations of NAA on rooting rate.average root number and average root length showed a trend of increasing first and then decreasincu,the suitable rooting basic medium was 1/2DKW with NAA 0.2mg/Lărooting rate was 62%(the highest was 74%),the average root number was 6.5,the average roof length was 2.9cm;(5)In the process of transplanting,the method of high pressure sterilization should be selcted to provide a better transition condition for the environmental transformation of plaritlets.the medium with the ratio of 1:2:1 between vermiculite,perlite and peat soil was the most suitable for seedling transplanting,94.4%of the plantlets survived. |
