Cornus waited which belongs to Corus genus of Cornaceae family has a lot of advantages,such as strong resistance, endurable to barren resistance, wide distribution and so on.It is not only excellent material for the production of timber, protection and ornamental tree, but also one of biodiesel oil tree species which can refine biodiesel. The cuttage propagation technique and root mechanism were studied in the paper and the results were as follows:1.Research on cuttage propagation techniqueStiff Stem Cuttage test results showed that: of the four factors taken,the primary and secondary effect between four factors is growth regulator> processing time take place> processing ear concentration. Under field conditions, the best combination is: choose the central branches cuttings and dip them in mixture of200mg/L NAA+IBA (1:1) growth regulator for2h.The result of orthogonal experiment in all optical atomizing shoots cottage of Cornus waited showed that five the primary and secondary effect between five factors is the processing time> hormone kind>cuttage medium>hormone concentration>cuttings length, the best combination is: choose16cm long cuttings and dip them in100mg/L NAA for an hour, then cutting in the river sand. The shoot cutting of Corus take root mainly with phloem and this belongs to Skin department took root. The result of randomized trialof shoot cutting showed: the optimum cuttings diameter is0.4-0.8cm; the optimum cuttings is from two years of biological mother tree; of the four cutting time chosen, the best is July22; cuttings with2-3leaves which are cut off half growthed better than those with all the leaves and those lack of leaves.2. Study on Physiological and biochemical techniqueThe results of the research show that the exogenous growth regulators can make the cuttings of the soluble contents more than the control so as to promote the cutting early to take root. And at the same time, in the process of generating roots of the hard branch and the delicate branch cuttings, the content of soluble sugar are in decline during the period of the callus occurrence and taking root. In the occurrence of the callus and adventitious roots, the starch degradation rate of the cuttings processed by hormone is far more quickly than the comparison. Study on the the activity of indoleacetic acid oxidase(IAAO) and peroxidase(POD) showed that both of them were all increasing markedly at the intial stage of cutting, but the the activity of POD decreased with the elongation of adventitious roots. The activity of IAAO treated by the growth regulator increased at first and then was on the trend of the lower, the activity of IAAO kept increasing in control group. Growth regulator processing IAAO activity increase at first on the trend of the lower.
