| Eye disease is a common oculopathy in the clinical practice of small animals,in which corneal ulcer occupies a large proportion in eye diseases,and the incidence of blindness is high.Platelet-rich fibrin(PRF)is a new type of platelet gel,as a new generation of platelet concentrates.PRF can release the growth factors slowly in tissue injury and play a role in promoting the repair of injury by its special three-dimensional structure.In addition,this technology has more advantages than platelet-rich plasma(PRP),which has the characteristics of operation fast,cheap and no need to add anything in it.In recent years,with the development of modern medicine,PRF technology has made progress in the research of soft tissue and hard tissue repair,and it has been applied in the field of bone regeneration medicine,oral and maxillofacial reconstruction,wound healing and so on.Test I Preparation and ultrastructural observation of platelet rich fibrinIn this study,12 healthy adult canines with body weight of 8-10kg were selected as the experimental canines.The PRF gel was prepared and its ultrastructure was scanned by electron microscopy(SEM)and transmission electron microscopy(TEM),on this basis,the morphological characteristics and biological characteristics were studied.It is observed that PRF is a porous network structure with a lot of trigeminal structure and a large number of platelets embedded in it.This structure is the basis of the structure of the growth factor and the premise of its biological characteristics.Test II Evaluation of the safety of autologous platelet rich fibrin in canine eyesIn this study,27 healthy adult canines with body weight of 8-10kg were selected as the experimental canines.The PRF membrane was covered on the cornea of the right eye,and the eyelids were sutured temporarily.Then,The PRF group was compared with blank group and negative control group.The safety was evaluated by observing the basic signs,the corneal inflammation score,the corneal ulcer score,the Schirmer test,the observation of ocular secretion,the hematology test,and the histopathological observation.The results showed that there was no obvious irritation and other adverse reactions on the corneal surface covered with PRF membrane.Test Ⅲ Establishment of corneal alkali burn model on canineIn this study,24 healthy adult canines with body weight of 8-10kg were selected as the experimental canines.The circular filter paper with a diameter of 10mm was dipped into lmol/L NaOH solution and then attached to the cornea of the right eye of the experimental canine.After contact with 40s,the surface of the ocular surface was washed with physiological saline immediately after 10min.The feasibility of this method was evaluated by gross observation,sodium fluorescein staining and Schirmer test.The results indicate that all of the models showed corneal color was gray white,corneal stroma edema,corneal epithelial damage and corneal fluorescein staining positive rate of 100%,that is,corneal alkali injury model was successfully produced.Test IV The evaluation of autologous Platelet-rich fibrin promote the repairing ability of the corneal ulcer in canineIn small animal clinic,we often encounter various causes of corneal ulcer.In this experiment,through the establishment of clinical model,clinical observation,corneal inflammation score,corneal ulcer score,Schirmer test,and measurement of corneal neovascularization and calculation of its area and pathological analysis,to evaluate the effect of autologous PRF on the repair of corneal ulcer in canine.The results showed that autologous PRF could repair the corneal alkali injury in canine,which is an effective new medical materials for ocular surface repair. |
