Transfer factor as a new type of immune enhancement agents,can improve the effect of vaccine immunization,strengthen body immunity,improve the animals free to prevention and control of infectious diseases in human and animal studies and clinical application is more and more widely.The purpose of this experiment was to prepare the spleen transfer factor of chicken,evaluate the quality of the spleen,and observe its influence on chicken immunity,so as to provide a basis for further clinical research.The chicken spleen transfer factor was prepared by using high speed mashing and low-temperature dialysis method.The properties of the products were examined by character,pH,ninhydrone and ammonium molybdate,the ratio of nucleic acid and polypeptide,protein,amino acid composition,etc.Biological characteristics such as biosafety,asepsis and macrophage phagocytosis of rats were examined.The results showed that the appearance of the transfer factor was yellowish and transparent liquid with slight fishy odor.The pH value was between 6.2 and 6.6,and the reaction of ninhydrin was purple and ammonium molybdate was precipitated.The ratio of nucleic acid and polypeptide is greater than 2,and the product contains no macromolecular protein.Glutamate content was the highest,with an average of 31.22 + 0.82μg/mL,and RSD of 3.72%;The content of methionine was lowest,with an average of 3.00±0.08μg/mL and RSD of 3.23%.The spirits,feeding of experimental group animal showed normal,the results of the autopsy were not abnormal;Non-bacterial growth,conforming to the requirements of biological products;Dose and high dose group of macrophage cell in abdominal cavity of mice percentage and index of chicken red blood cells were significantly higher than that of control group(P < 0.05),low dose group and the blank group difference was not significant(P > 0.05).Test randomly choose 200 chickens into Ⅰ,Ⅱ,Ⅲ,Ⅳ,Ⅴ five groups,each group of 40.Ⅰ group normal vaccine;Ⅱ,Ⅲ,Ⅳ group on the basis of the normal immune,each one separately intraperitoneal injection of 0.8 mL and 1.0 mL and 1.2 mL,Ⅴ control group,not immune,and intranasal approach during the age of 28 artificial ND virus infection.The young chicken was inoculated with a nasal drip in the 7th day of age.At age 14,the inoculation method of the method of drinking water was inoculated with polyvalent poisoning.On 21 days of age,the new Newcastle disease was inoculated with the new Newcastle disease.The test chicken was adopted on the net,and the experimental animals were prepared in the same way as NRC(1994),and the test period was 42 d.The contents included: average weight and weight gain,chicken mortality,chicken immune organ growth index,T and B lymphocyte conversion rate,HI antibody titer.,according to the results of 7-28 d group Ⅲ chicken the most weight,average weight is 1350.6 g,the average daily gain of 56.6 g,28-42 d group Ⅲ chicken the most weight,average weight is 2286.3 g,the average daily gain of 66.8 g,compared with group Ⅰ comparison and Ⅴ,difference was significant(P < 0.05);After 42 d,group Ⅲ broiler mortality rate was only 3.3%,group Ⅴ broiler mortality increase sharply to 40.0%,autopsy showed typical Newcastle disease pathogenesis characteristics;Compared with GroupⅠ,42 d Group Ⅲ poultry immune organs index were significant difference(P < 0.05),a group of Ⅳ chicken spleen index was not significant difference(P > 0.05),thymus index significantly difference(P < 0.05),group Ⅱ the immune organ index difference was not significant(P > 0.05);Compared with group Ⅴ,Ⅰ to group Ⅳ poultry immune organs index were significant difference(P < 0.05);Compared with GroupⅠ,42 d Group Ⅲ Ⅳ chicken T,B lymphocyte transformation were significant difference(P < 0.05),group Ⅱ was no significant difference(P > 0.05);Compared with group Ⅴ,poultry groups in terms of T,B lymphocyte transformation were significant difference(P < 0.05);When compared with groups Ⅰ,14 d groups of HI antibody differences antibody concentration was not significant(P > 0.05);35 d and 28 d,42 d group Ⅲ and Ⅳ Ⅰ significant difference(P < 0.05),group Ⅱ and Ⅰ difference was not significant(P > 0.05).14-42 d,each group compared with group Ⅴ,HI antibody degrees were significantly difference(P < 0.05).The above results show that the preparation of chicken spleen transfer factor security was good,can reduce broiler mortality,promote the growth of broilers,and increases chicken immune organ index,promoting the transformation of T 、 B lymphocytes and HI antibody,promote cellular immunity and humoral immunity function of broilers.This experiment showed that injection of high dose effect of transfer factor was better than the low dose,immune effect and dose have certain dependency,but not the higher the better,each 1 mL injection quantity was best.
