Transfer factor (transfer factor, TF) is produced by the T lymphocyte havingimmunological competence in the white blood cells, its molecular weight is small andchemical nature is small peptide and oligonucleotide complexes. This experiment through theacquisition of healthy pigs’ spleen, non-specific TF were prepared by dialysis method, TFwere detected, results show:the protein reaction is negative and the content of polypeptide isequal to0.615mg/ml, OD260/OD280is2.35, DNA content determination is45.2(ug/ml). Theresults and data reported TF’ quality control standard is consistent. Through biological test forthe detection of TF, results show: TF is non-toxic, no heat, no allergy, macrophage phagocyticphenomenon obviously, E-rosette flowering rate increased, indicating that TF has the activityof immune function. This study uses the ELISA, flow cytometry, automatic blood cellanalyzer to test pigs with TF vaccine injection, different time change detection of swinefoot-and-mouth disease, circovirus antibody, the changes of lymphocyte number and ratio,changes of T cell subsets, cytokine that has a key role in immune response regulation (IL-2and IFN-γ) change secretion. On this basis, we see the feasibility of using porcine circovirusvaccine dose and the necessity of emergency prevention of swine foot-and-mouth diseasewith TF.This research consists of four parts as follows:Part One: Production of transfer factorThe production process of transfer factor (TF): Pig spleen was pounded to homogenateand dialysised after frozen, melt, freeze-thaw for6-8times. When the physical-chemicalproperties and biological characteristics of the dialysis fluid met the requirements, the dialysisfluid was qualified.Part Two: Immune-enhancing effect of Transfer FactorTransfer Factor, which met the requirements, as a safe, economic, easy to use andefficient vaccine adjuvant showed great immune-enhancing effect, just as follows: vaccinatedwith the FMD vaccine and the original TF3ml, the next day injected3ml transfer factoragain,could improve the content of FMD antibody, which was from70ng/L of7d injected to300ng/L of35d; could improve the ratio of blood lymphocytes, lymphocyte counts; couldimprove the amount of IFN-γ; time of secreting IFN-γ was short and the highest was after7dinjected in this test;could extend the time of lymphocytes secreting IL-2and the highest wasafter28d injected. Part three: Transfer factor can be used as vaccine adjuvants to reduce the usage ofvaccines12-day-old piglets are immunized1.2ml porcine circovirus vaccine with3ml transferfactor by intramuscular injection and inject another3ml transfer factor the next day.Then7-35days period of time, it achieve the same effect of immunizing2ml porcine circovirusvaccine.So transfer factor be used as a vaccine adjuvantdemonstrate huge immune-enhancingeffect. Compared with12-day-old piglets being immunized0.9ml porcine circovirus vaccinewith3ml transfer factor by intramuscular injection and another3ml transfer factor the nextday.It’s effect is less than immunizing2ml porcine circovirus vaccine by indexes, and thedifference was significant, statistically significant(P <0.05).Part four:Transfer factor can improve the porcine peripheral blood CD4~+cell numbersand CD4~+/CD8~+ratio.Immunize1.2ml swine foot-and-mouth disease vaccine with3ml transfer factor byintramuscular injection and inject another3ml transfer factor the next day. Control group onlyimmunize2ml swine foot-and-mouth disease vaccine.7,14,21-day-old test results showedthat T lymphocyte subsets CD4~+T cell percentage and CD4~+/CD8~+ratio are the higheston14-day-old,followed by decreased slightly.T cells CD4~+and CD4~+/CD8~+ratio of theexperimental group are higher than control group on the same age.Significant differences inthe experimental group and the control group was statistically significant (P <0.05).Sotransfer factor can improve the porcine peripheral blood CD4~+cell numbers and CD4~+/CD8~+ratio.
