Konjac glucomanan(KGM)could obtain vastly from Amorphophallus.It displays various properties in several aspects,such as hydrophilicity,gelation,and antibacterial.Additionally,it is widely used in medicine,chemical,textile,paper,food as well as other fields.A.muelleri is a potential popularized species,and its corms contain lots of glucomannan.However,studies about A.muelleri were rare.In this paper,the accumulation and influential factors of glucomannan during corms enlarging were studied.At first,the dynamic changes of morphological and quality indexes during corms enlarging were analyzed,and the contents of glucomannan and starch were also measured.Subsequently,the contents of glucomannan and starch were analyzed using different temperature and hormones treating.Additionally,starch synthesis-related gene,AmAGP,were detected during the corms enlarging in A.muelleri.At last,the transcript levels of AmAGP were also detected using the treatment of different temperature and hormones.The main results are as follows: 1.Morphological and quality indicators were analyzed during corm enlaring in A.muelleri.Accompany with the enlargement of corm,the growth rate of roots were increased slowly at first and then rapidly after;the increasement of petiole diameter were smooth;the hight of plants and fresh corm were increased rapidly at first,and then were smoothly.The contents of konjac klucomanan and starch during the corm enlarging were measured in corms,the results showed that starch were increased vastly at first and decrased after,konjac klucomanan were increased largely at first and smoothly then.2.Contentsof glucomannan were measured in corms using different temperature treatments.In this study,the seedlings of A.muelleri were treated using different temperatures(25?,30?and 35?),and the contents of glucomannan were detected.The results showed that 35°C was the favorable temperature for the accumulation of glucomannan in the corms.3.Contents of glucomannan were measured in corms under the treatments using different hormones.In this study,the plants of A.muelleri were treated using three hormones(indole acetic acid,thidiazuron,and salicylic acid),and the contents of glucomannan and starch were detected in corms.The results showed that indole acetic acid promoted the accumulation of glucomannan in corms.4.Transcript level of AmAGP were detected in corms during enlargement of A.muelleri.The results showed that the expression level of AmAGP gene was higher at 120 days after sowing,and followed by 90 days and 150 days after sowing,respectivly.5.Expression level of AmAGP gene were analysed in corms using the treatments of different temperature and hormone treatments.Under the treatments using different temperatures and hormones,the expression level of AmAGP were detected in corms.The results showed that 25°C and indole acetic acid induced the expression of AmAGP.
